参芪复方调控PI3K/AKT/mTOR信号通路改善糖尿病GK大鼠胰岛功能减少血糖波动的实验研究 被引量:6

摘要 目的探讨参芪复方调控PI3K/AKT/mT OR信号通路,对糖尿病GK大鼠胰岛功能及血糖波动的影响。方法建立由高脂饲料喂养的糖尿病GK大鼠模型。实验分4组:正常组,模型组,中药组,西药组。于灌胃后每周1次测一日内8:00,10:00,14:00,16:00,18:00时血糖水平,并用血糖稳定性指标(MBG、SDBG、LAGE)量化各组血糖稳定性;检测胰岛素、胰高血糖素,TUNEL法检测胰岛β细胞凋亡,并运用实时荧光定量PCR检测胰岛β细胞凋亡相关基因表达水平。结果参芪复方组可减少大鼠血糖波动,减少胰岛β细胞凋亡,调控胰岛β细胞凋亡相关的PI3K/AKT/mT OR信号通路基因。结论具有养阴益气活血功效的参芪复方可调控PI3K/AKT/mT OR信号通路,减少胰岛β细胞凋亡,改善胰岛功能,达到调节血糖稳态。 Objective To explore the mechanism of Shenqi Compound Formula on the control of PI3 K/AKT/mT OR signal pathway to improve islet function and reduce blood sugar fluctuation in diabetic GK rats. Methods We established a rat diabeticmodel with high-fat diet. The rats were randomly divided into normal group,model group,traditional Chinese medicine group,Western medicine group,a total of 4 groups. Starting from the gavage administration,we measured the blood glucose levels once a weekat 8: 00,10: 00,14: 00,16: 00,18: 00. The blood glucose stability was quantified with the indexes of MBG,SDBG and LAGE.The insulin,glycotin and TUNEL were detected for apoptosis of islet beta cells,and gene expression levels associated with apoptosis of islet beta cells were detected by Real-time fluorescence quantitative PCR. Results The traditional Chinese medicine group can reduce the blood sugar fluctuation of rats,reduce the apoptosis of islet beta cells,and regulate the PI3 K/AKT/mT OR signaling pathway genes associated with islet beta cell apoptosis. Conclusion Shenqi Compound Formula which has the function of supplementing Qi and Yin and promoting blood circulation may achieve the function of regulating the PI3 K/AKT/mT OR signal pathway,reducing the apoptosis of islet beta cells,improving islet function and regulating blood sugar homeostasis.
