摘要
目的观察针刺对颅脑损伤(TBI)大鼠海马组织单羧酸转运蛋白MCT2、MCT4表达的影响,探讨针刺促TBI大鼠康复的海马能量代谢机制。方法33只SD大鼠随机分为模型组11只,针刺组12只,空白组10只。模型组和治疗组采用Feeney自由落体撞击法制备TBI模型大鼠,造模12 h后治疗组进行针刺干预,选取"人中""百会""内关""足三里"进行针刺,1次/d,共治疗10次。采用神经功能评价、行走功能评价、平衡功能评价以及肢体回缩力测试对各组大鼠进行行为学评价,Western blot和免疫组化方法检测大鼠海马组织MCT2、MCT4蛋白表达。结果与空白组相比,模型组大鼠神经功能显著下降(P<0.01),平衡功能评分显著升高(P<0.01),行走耗时显著延长(P<0.01);针刺治疗后,针刺组大鼠神经功能改善不具有统计学意义(P>0.05),平衡功能和行走功能均得到显著提升(P<0.05,P<0.05)。Western blot检测显示,与空白组相比,模型组大鼠海马组织MCT2、MCT4蛋白表达显著降低(P<0.01),针刺治疗后,MCT2、MCT4蛋白表达显著升高(P<0.01,P<0.05);免疫组化检测显示,与空白组相比,模型组大鼠海马组织MCT2、MCT4蛋白表达显著降低(P<0.01),针刺治疗后,针刺组大鼠MCT2蛋白表达显著升高(P<0.05),MCT4蛋白表达升高不具有统计学意义(P>0.05)。结论针刺可以改善TBI大鼠平衡和行走功能,促进其神经功能康复,可能与促进海马组织MCT2、MCT4表达,调控神经元能量转运代谢有关。
Objective To observe the effect of acupuncture on the expressions of MCT2 and MCT4 in hippocampus of traumatic brain injury( TBI) rats so as to explore the energy metabolic mechanism of acupuncture in neurorehabilitation. Methods Thirty-three rats were randomized into model group( n = 11),acupuncture group( n = 12) and control group( n = 10). Rats in model group and acupuncture group were established TBI model by using Feeney method. After 12 hours recovery,the acupuncture treatment on "Renzhong( DU26) ","Baihui( DU20) ","Neiguan( PC6) "and "Zusanli( ST36) "was applied once a day,lasting for10 days in total. The neural function of rats was detected by using neurological function evaluation,walking function evaluation and balance function evaluation. The expressions of MCT2 and MCT4 in hippocampus were detected by Western blot and immunohistochemistry. Results Compared with that of the control group,the neurological function of rats in model group was significantly decreased( P < 0. 01). The balance function evaluation score rose( P < 0. 05) and time of walking prolonged( P < 0. 01). After acupuncture treatment,compared to the model group,rats in acupuncture group showed an improvement in balance function( P <0. 05) and walking function( P < 0. 05),but no significantly differences were observed in neurological function evaluation( P >0. 05). The Western blot results suggested that the expression levels of MCT2 and MCT4 were significantly lower in the model group than those in the control group( P < 0. 01). Compared with those of the model group,the expression levels of MCT2 and MCT4 in the acupuncture group were significantly higher( P < 0. 01,P < 0. 05). The immunohistochemistry results showed expression levels of MCT2 and MCT4 in model group were significantly lower compared with those of the control group( P < 0. 01). Compared with those of the control group,the expression of MCT2 in acupuncture group significantly increased( P < 0. 05),and no significantly differences were found in the expression of MCT4 in the acupuncture group( P > 0. 05). Conclusion Acupuncture can improve the balance and walking functions,and promote the expressions of MCT2 and MCT4 in hippocampus of TBI rats. The improvement of neurological function may be related to the regulation of neuronal energy metabolism.
