摘要
长链非编码RNA(LncRNA)是长度大于200个核苷酸的非蛋白质编码转录物,近几年来的研究表明,LncRNA上具有结合microRNA(miRNA)的部分互补序列,这些互补序列通过竞争性结合miRNA,从而抑制miRNA对靶基因mRNA的表达作用。也就是说,LncRNA能够作为一种竞争性内源RNA(ceRNA),通过海绵吸附的方式与miRNA竞争性结合,从而阻止miRNA同其靶mRNA结合,进而调控蛋白质的表达水平。在临床上,肝细胞癌(HCC,简称肝癌)通常被诊断为最致命的恶性肿瘤之一,其在全球癌症相关死亡中居于前列,肝癌因其发病机制的复杂性以及对化疗药物的耐药性,使得HCC的诊断和治疗以及预后仍然不能令人满意。随着研究的不断进展,人们发现LncRNA除了可以直接参与基因的表达调控外,还能够作为一种竞争性内源RNA(ceRNA)与miRNA相互作用,从而在HCC的进展中参与调控细胞增殖、死亡以及侵袭与转移等过程。该文从LncRNA海绵吸附miRNA发挥生物学功能这个方面,归纳总结相关LncRNA在肝癌发生发展中的作用及其与中医药干预机制的研究进展,以此为肝癌的诊断与治疗提供新思路。
Long-chain non-coding RNA( LncRNA) is a non-protein-coding transcript longer than 200 nucleotides.Studies in recent years have shown that LncRNA has partially complementary sequences that bind to microRNA( miRNA). These complementary sequences bind miRNA competitively,thereby inhibiting the expression of miRNA on target gene mRNA. That is to say,LncRNA can be used as a competitive endogenous RNA( ceRNA),and it can competitively bind to miRNA through sponge adsorption,thereby preventing miRNA from binding to its target mRNAandregulating protein expression level. Clinically,hepatocellular carcinoma( HCC,liver cancer for short) is usually diagnosed as one of the most deadly malignant tumors,and it ranks in the forefront of cancer-related deaths worldwide. Liver cancer is due to the complexity of its pathogenesis and resistance to chemotherapy drugs,making the diagnosis and treatment of HCC and prognosis still unsatisfactory. With the continuous progress of research,it is found that LncRNA can not only directly participate in the regulation of gene expression,but also can interact with miRNA as a competitive endogenous RNA( ceRNA),thereby participating in the regulation of cell proliferation and death in the progress of HCC and the process of invasion and transfer. This article summarized the role of LncRNA sponge in adsorbing miRNA to play a biological role,and summarized the role of LncRNA in the development of liver cancer and the research progress of traditional Chinese medicine intervention mechanisms,in order to provide new ideas for the diagnosis and treatment of liver cancer.
