微小RNA-139-5p对AS患者血管平滑肌细胞增殖和迁移作用的研究 被引量:2

摘要 目的:检测微小RNA-139-5p(miR-139-5p)在动脉粥样硬化(AS)患者中的表达情况,分析miR-139-5p的临床价值及对人血管平滑肌细胞(HVSMCs)生物学功能的调控作用。方法:采用荧光定量PCR技术检测112例AS患者(AS组)和78例健康志愿者(对照组)血清miR-139-5p的表达情况,用受试者工作特征曲线(ROC曲线)评估miR-139-5p对AS的诊断价值。采用CCK-8实验和Transwell实验分析miR-139-5p对HVSMCs增殖和迁移能力的影响。结果:与对照组相比,AS组患者miR-139-5p的表达明显下调(P<0.001),同时AS组患者血清miR-139-5p的表达水平与颈动脉内中膜厚度(CIMT)呈负相关(r=-0.7579,P<0.001)。miR-139-5p诊断AS的ROC曲线下面积(AUC)为0.888,具有较高敏感性(86.6%)和特异性(84.6%)。miR-139-5p低表达会促进HVSMCs的增殖和迁移能力,相反miR-139-5p过表达会显著地抑制HVSMCs的增殖和迁移能力(P<0.001)。结论:miR-139-5p的低表达可能是AS的潜在诊断标志物,过表达miR-139-5p可能通过抑制HVSMCs的增殖和迁移发挥改善AS的潜能。 Objective:To assess the clinical value of MicroRNA-139-5p(miR-139-5p)and analyze its regulatory effect on the biological behavior of vascular smooth cells(HVSMCs)by detecting the expression of miR-139-5p in atherosclerosis(AS)patients.Methods:The expression of serum miR-139-5p in 112 AS patients(AS group)and 78 healthy controls(control group)were detected by fluorescence quantitative PCR.Receiver operating characteristic(ROC)curve was used to evaluate the diagnostic value of miR-139-5p in AS.The effect of miR-139-5p on HVSMCs proliferation and migration was analyzed by CCK-8 assay and Transwell assay.Results:The expression of miR-139-5p in AS group was significantly down-regulated compared with the healthy control group(P<0.001).Meanwhile,the expression level of miR-139-5p in serum of AS group was negatively correlated with carotid intima-media thickness(CIMT)(r=0.7579,P<0.001).The area under the ROC curve(AUC)was 0.888,with a high sensitivity(86.6%)and specificity(84.6%).In addition,the low expression of miR-139-5p promoted the proliferation and migration of HVSMCs,while the overexpression of miR-139-5p significantly inhibited the proliferation and migration of HVSMCs(P<0.001).Conclusion:The decreased expression of miR-139-5p may be a potential diagnostic marker of AS,and overexpression of miR-139-5p may improve AS by inhibiting the proliferation and migration of HVSMCs.
