摘要 目的探究不同浓度的维生素E注射到衰老大鼠的海马体对其空间记忆功能的影响。方法将32只SD大鼠随机分为4组,每组各8只。造模后进行分组,实验对照组PBS溶液(甲)、12.5%浓度维生素E组(乙)、25%浓度维生素E组(丙)和50%浓度维生素E组(丁)。先用10%D-半乳糖制造衰老大鼠模型,造模后在大鼠脑立体定位仪下分别对甲、乙、丙、丁组大鼠海马体注入PBS、12.5%浓度维生素E、25%浓度维生素E和50%浓度维生素E。脑注射并缝合术后第4天,对大鼠进行Morris水迷宫实验,观察大鼠定向游泳潜伏期和滞留期的长短以及穿越站台的次数。结果4组大鼠情况如下,乙组大鼠定向游泳潜伏期较甲组大鼠短(P<0.05);丙组大鼠定向游泳潜伏期较乙组大鼠长(P<0.05);丁组大鼠定向游泳潜伏期较乙组大鼠长(P<0.05);丙组大鼠定向游泳潜伏期比丁组短(P<0.05);滞留期时长从长到短依次为乙组、丙组、丁组、甲组(P<0.05);穿越平台数数量从多到少依次为乙组、丙组、丁组、甲组(P<0.05)。结论维生素E可改善衰老大鼠海马体的空间学习记忆功能,导致其定向游泳潜伏期缩短,滞留期变长,穿越平台数增多;且较低浓度维生素E的改善效果较好,在一定阶梯浓度中,12.5%浓度的维生素E改善效果最佳。 Objective To investigate the effects of different concentrations of vitamin E injected into the hippocampus of aging rats on their spatial memory function.Methods 32 SD rats were randomly divided into 4 groups,with 8 rats in each group.Modeling was performed after the grouping,and the experimental group was treated with PBS solution(A),12.5%vitamin E(B),25%vitamin E(C),and 50%vitamin E(D).The aging rat model was firstly made with 10%D-galactose.After modeling,PBS,12.5%vitamin E,25%vitamin E and PBS,and 50%concentration of vitamin E were injected into the group A,B,C and D,respectively.On the third day after the injection of vitamin E in the brain and the suture,the rats were subjected to a Morris water maze test to observe the length of the directional swimming latency and retention period of the rats and the number of crossing the platform.Results The four groups of rats were as follows.The directional swimming latency of group B was shorter than that of group A(P<0.05).The directional swimming latency of group C was longer than that of group B(P<0.05).The directional swimming latency of the rats in the D group was longer than that in the B group(P<0.05).The directional swimming latency of the rats in the C group was shorter than that of the D group(P<0.05);the duration of the detention period was from B to B.Group,C group,D group,A group(P<0.05);the number of crossing platforms was B group,C group,D group,and A group(P<0.05).Conclusion Vitamin E can improve the spatial learning and memory function of the hippocampus of aging rats,resulting in shortened directional swimming latency,longer retention period and more number of crossing platforms.The lower concentration of vitamin E improves better,in a certain step concentration.12.5%concentration of vitamin E improved best.
