含酒大黄与醋大黄的大承气汤中10个活性成分含量测定研究 被引量:3

摘要 目的 HPLC法同时测定酒大黄与醋大黄的大承气汤中10个活性成分含量。方法色谱柱为Agilent Zorbax SB-C18(5μm,4.6 mm×250 mm),流动相为乙腈-0.1%磷酸水溶液,梯度洗脱,流速为1 mL·min^-1,检测波长280 nm和254 nm,柱温30℃的条件下,测定含酒大黄与醋大黄的大承气汤中10个活性成分的含量,并分析其含量差异。结果 10条标准曲线在线性范围内均呈良好线性关系;方法学考察达到试验要求,含酒大黄与醋大黄的大承气汤中的10个活性成分含量均有不同程度的变化。结论含酒大黄与醋大黄的大承气汤中10个活性成分含量均有不同程度的变化,这些变化与酒大黄和醋大黄配伍的大承气汤作用相对应,且大黄蒽醌类成分含量的变化在一定程度上对大承气汤的功效存在着一定的影响和共性关系。 Objective Simultaneous determination of 10 active ingredients in Dachengqi Decoction of rhubarb and vinegar rhubarb by HPLC. Methods The column was Agilent Zorbax SB-C18(5 μm,4.6 mm×250 mm),the mobile phase was acetonitrile-0.1% phosphoric acid aqueous solution,gradient elution,flow rate was 1 mL·min^-1,detection wavelength was 280 nm and 254 nm,column temperature was 30 ℃. Under the conditions,the content of 10 active ingredients in Dachengqi Decoction of wine rhubarb and vinegar rhubarb was determined,and the difference in content was analyzed. Results The 10 standard curves showed a good linear relationship in the linear range;the methodological investigation reached the test requirements,and the content of 10 active ingredients in Dachengqi Decoction of rhubarb and vinegar rhubarb varied to varying degrees. Conclusion Both wine rhubarb and vinegar rhubarb have different degrees of changes in the content of 10 active ingredients in Dachengqi Decoction. These changes correspond to the effect of Dachengqi Decoction in combination with rhubarb and vinegar rhubarb,and the content of rhubarb mites is changed. To a certain extent,there is a certain influence and common relationship on the efficacy of Dachengqi Decoction.
