摘要
目的利用柱前衍生HPLC法测定麻黄粗多糖中4种单糖的含量。方法采用PMP柱前衍生的方法,色谱柱Diamonsil C18色谱柱(250×4.6mm, 5μm)流动相100mmol·L^-1(p H值为6.7)磷酸缓冲盐-乙腈等度洗脱,流速0.8m L·min^-1,检测波长245nm。结果在优化后的分析条件下,平均回收率范围在98%~102%,且线性良好。结论该方法准确,简便,适合于麻黄总糖中单糖含量的精密测定。
AIM HPLC with pre-column derivatization method was used to determine the content of 4 monosaccharides in Ephedra polysaccharide.METHODS By the PMP pre-column derivatization method,the HPLC separation was achieved on a Diamonsil C18(250×4.6mm,5μm)column.The mobile phase used 100mmol·L-1(pH=6.7)phosphate buffered saline-acetonitrile with isocratic elution and the flat raw is 0.8mL·min-1,UV detection at 245nm.RESULTS Under the established analytical condition,the average recoveries were 98%~102%,and had good linearity.CONCLUSION The method is accurate,simple,convenient and suitable for the determination of monosaccharides in the Exphedra polysaccharides.
