摘要
目的研究调气消积汤对Lewis肺癌小鼠的抑瘤作用及对血清TNF-α水平的影响。方法将60只Lewis肺癌小鼠随机分为4组,每组15只。肿瘤组给予蒸馏水灌胃,调气消积汤组给予调气消积汤灌胃,顺铂组给予顺铂药液灌胃,调气消积汤+顺铂组给予调气消积汤和顺铂灌胃,连续灌胃12 d。观察4组小鼠的死亡数量、反应性、体毛、大便及进食量,末次灌胃24 h后处死小鼠,称量瘤质量,计算抑瘤率,检测血清肿瘤坏死因子-α(TNF-α)水平。结果实验过程中顺铂组死亡1只,其他各组没有发生死亡。调气消积汤组小鼠反应性较灵敏,体毛有光泽,大便成形,其他组反应迟钝,体毛无光泽,大便不成形;调气消积汤组小鼠的进食量显著高于其他组(P均<0.05),顺铂组小鼠的进食量显著低于其他组(P均<0.05),调气消积汤+顺铂组小鼠的进食量与肿瘤组比较差异无统计学意义(P>0.05)。各给药组小鼠的移植瘤质量均显著低于肿瘤组(P均<0.05),顺铂组、调气消积汤+顺铂组显著低于调气消积汤组(P均<0.05),顺铂组、调气消积汤+顺铂组比较差异无统计学意义(P>0.05);顺铂组、调气消积汤+顺铂组抑瘤率显著高于调气消积汤组(P均<0.05),且调气消积汤+顺铂组高于顺铂组(P<0.05)。顺铂组、调气消积汤+顺铂组血清TNF-α水平显著高于肿瘤组和调气消积汤组(P均<0.05),顺铂组与调气消积汤+顺铂组比较、肿瘤组与调气消积汤组比较差异均无统计学意义(P均>0.05)。结论调气消积汤能够改善Lewis肺癌小鼠的生存状况,对肿瘤有抑制效果,与顺铂合用抑瘤作用更为显著,但其对血清TNF-α水平没有明显影响。
Objective It is to study the anti-tumor effect of Tiaoqi Xiaoji Decoction on Lewis lung cancer mice and its effect on serum TNF-αlevel.Methods Sixty Lewis lung cancer mice were randomly divided into 4 groups:tumor group,Tiaoqi Xiaoji Decoction group,cisplatin group and Tiaoqi Xiaoji Decoction+cisplatin group,15 mice in each group.The tumor group was given distilled water by gavage,the Tiaoqi Xiaoji Decoction group was given Tiaoqi Xiaoji Decoction by gavage,the cisplatin group was given cisplatin liquid by gavage,and the Tiaoqi Xiaoji Decoction+cisplatin group was given Tiaoqi Xiaoji Decoction and cisplatin by gavage,all the groups were continuously treated for 12 days.The number of deaths,reactivity,body hair,stool and food intake of the mice of the 4 groups were observed.The mice were sacrificed in 24 hours after the last gavage.The tumor mass was weighed,the tumor inhibition rate was calculated,and the level of serum tumor necrosis factor-α(TNF-α)was detected.Results During the experiment,one animal died in the cisplatin group,and no death occurred in the other groups.The mice in the Tiaoqi Xiaoji Decoction group were more sensitive,with shiny body hair and shaped stools,while the mice in the other groups were slow to respond,with dull body hair and unshaped stools.The mice in the Tiaoqi Xiaoji Decoction group had significantly higher food intake than that of the mice in the other groups(P<0.05),the food intake of mice in the cisplatin group was significantly lower than that of the other groups(all P<0.05),the food intake of mice in the Tiaoqi Xiaoji Decoction+cisplatin group was not significantly different from that of the tumor group(P>0.05).The weight of transplanted tumors of mice in each administration group was significantly lower than that of the tumor group(all P<0.05),and the weight of the cisplatin group,Tiaoqi Xiaoji Decoction+cisplatin group was significantly lower than that of the Tiaoqi Xiaoji Decoction group(all P<0.05),there was no significant difference between the cisplatin group and the Tiaoqi Xiaoji Decoction+cisplatin group(P>0.05).The tumor inhibition rate in the cisplatin group,Tiaoqi Xiaoji Decoction+cisplatin group was significantly higher than that in the Tiaoqi Xiaoji Decoction group(P<0.05),and the Tiaoqi Xiaoji Decoction+cisplatin group was higher than the cisplatin group(P<0.05).The levels of serum TNF-αin the cisplatin group,Tiaoqi Xiaoji Decoction+cisplatin group were significantly higher than those in the tumor group and Tiaoqi Xiaoji Decoction group(P<0.05),there was no significant difference in the levels of serum TNF-αbetween cisplatin group and Tiaoqi Xiaoji Decoction+cisplatin group,between the tumor group and the Tiaoqi Xiaoji Decoction group(all P>0.05).Conclusion Tiaoqi Xiaoji Decoction can improve the survival status of Lewis lung cancer mice,and has an inhibitory effect on tumors,the effect is more significant when combined with cisplatin,but it has no significant effect on serum TNF-αlevel.
