LncRNA NNT-AS1、HMGB1在膀胱癌组织中的表达水平及其临床意义 被引量:3

Expressions and clinical significance of LncRNA NNT-AS1 and HMGB1 in bladder cancer

摘要 目的探讨长链非编码RNA(LncRNA)烟酰胺核苷酸反义转氢酶RNA1(NNT-AS1)、高迁移率族蛋白B1(HMGB1)在膀胱癌组织中的表达水平及其与预后关系。方法选取广州医科大学附属中医医院2010年1月-2014年12月诊治的膀胱癌患者的癌组织及对应癌旁正常组织各73例标本进行研究。采用实时荧光定量PCR(qRT-PCR)法检测膀胱癌癌组织及对应癌旁正常组织LncRNA NNT-AS1、HMGB1表达水平;分析膀胱癌癌组织LncRNA NNT-AS1、HMGB1表达水平与膀胱癌患者临床病理特征、预后的关系;Pearson法分析LncRNA NNT-AS1表达水平与HMGB1相关性。结果膀胱癌患者癌组织LncRNA NNT-AS1、HMGB1表达水平均明显高于癌旁正常组织(t=8.769、9.423,P均<0.05);膀胱癌癌组织LncRNA NNT-AS1、HMGB1表达水平与膀胱癌患者TNM分期、淋巴结转移、分化程度相关(P<0.05);膀胱癌癌组织LncRNA NNT-AS1表达水平与HMGB1呈正相关(r=0.394,P<0.05);膀胱癌患者LncRNA NNT-AS1、HMGB1高表达组术后60个月生存率均明显低于LncRNA NNT-AS1、HMGB1低表达组(P均<0.05)。结论 LncRNA NNT-AS1、HMGB1在膀胱癌癌组织中均呈高表达,两者均与膀胱癌患者预后有关,有助于评估膀胱癌患者预后情况。 Objective To investigate the expressions of long non-coding RNA(LncRNA) nicotinamide nucleotide transhydrogenase-antisense RNA1(NNT-AS1) and high mobility group box-1 protein(HMGB1) in bladder cancer and their relationships with prognosis.Methods 73 specimens of cancer tissues from bladder cancer patients diagnosed and treated in our hospital from January 2010 to December 2014 and corresponding normal tissues adjacent to cancer were selected for research.The expression levels of LncRNA NNT-AS1 and HMGB1 in bladder cancer cancer tissues and corresponding normal tissues were detected by real-time fluorescent quantitative PCR(qRT-PCR) method;the relationships between the expressions of LncRNA NNT-AS1,HMGB1 in bladder cancer cancer tissue and the clinicopathological features,prognosis of bladder cancer patients were analyzed;and the correlation between the expression levels of LncRNA NNT-AS1 and HMGB1 was analyzed by Pearson method.Results The expression levels of LncRNA NNT-AS1 and HMGB1 in cancer tissue of bladder cancer patients were significantly higher than those in adjacent normal tissues(P<0.05);the expression levels of LncRNA NNT-AS1 and HMGB1 in bladder cancer tissues were correlated with TNM stage,lymph node metastasis and differentiation in bladder cancer patients(P<0.05);the expression of LncRNA NNT-AS1 in bladder cancer tissues was positively correlated with HMGB1(P<0.05).The 60-month survival rate of bladder cancer patients with high expressions of LncRNA NNT-AS1 and HMGB1 was significantly lower than that of low expressions of LncRNA NNT-AS1 and HMGB1(P<0.05).Conclusion LncRNA NNT-AS1 and HMGB1 were highly expressed in cancer tissues of bladder cancer patients;both of them were related to the prognosis of bladder cancer patients,which could be helpful to evaluate the prognosis of bladder cancer patients.
