摘要
目的研究酒萸肉鉴别检测方法,为规范饮片生产及加强监管力度提供依据。方法以5-羟甲基糠醛(5-hydroxymethylfurfural,5-HMF)为对照品,薄层色谱法初筛,对山萸肉、酒萸肉进行快速有效的区分;采用C18柱(50 mm×2.1 mm,1.8μm),流动相为0.005 mol/L乙酸铵溶液-乙腈,流速0.2 mL/min梯度洗脱,波长240nm,超高效液相色谱法检测,并用液质联用验证。结果在薄层色谱法(thin layer chromatography,TLC)、超高效液相色谱法(ultra performance liquid chromatography,UPLC)和液相色谱-质谱联用法(liquid chromatography-mass spectrometry,LC-MS)佐证结果一致的情况下,所建立的薄层色谱法、液相色谱方法能有效区别山萸肉、酒萸肉。结论该方法快速、准确、专属性好,可作为酒萸肉鉴别检测方法,同时可为酒萸肉质量控制提供方法。
Objective To investigate the method for identification and detection of processed Fructus corni, and provide basis for standardizing the production and strengthening the supervision. Methods The thin layer chromatography(TLC) identification condition of crude and processed Cornus officinalis fruit was studied with 5-hydroxymethylfurfural as reference standard. A C18 column(50 mm×2.1 mm, 1.8 μm) was adopted for the determination, which was maintained at 30 ℃ throughout the analysis. The mobile phase was composed of 0.005 mol/L ammonium acetate and acetonitrile at a flow rate of 0.2 mL/min. The UV detector was set at 240 nm. The detection was performed by ultra performance liquid chromatography(UPLC) and validated by liquid chromatography-mass spectrometry(LC-MS). Results The results of TLC was consistent with those by UPLC and LC-MS. It could be used as an identification method of crude and processed Fructus corni. Conclusion The method is rapid, accurate and specific, which can be used as the identification and detection method of Fructus corni and provide a method for the quality control of Fructus corni.
