摘要
目的研究牛蒡子苷元(arctigenin,ARG)对鼻咽癌5-8F细胞裸鼠移植瘤生长影响及其分子机制。方法建立鼻咽癌5-8F细胞裸鼠移植瘤模型,随机分为对照组、10μg/g ARG组和20μg/g ARG组。实验组按体重分别给予10μg/g和20μg/g牛蒡子苷元,对照组则给予等体积1640培养液。每天腹腔注药,持续2周。绘制生长曲线,计算抑瘤率;Real-time PCR法及Western blot法检测mRNA及蛋白表达水平。结果ARG能抑制鼻咽癌5-8F细胞裸鼠移植瘤生长;与对照组相比,10μg/g ARG组和20μg/g ARG组表皮生长因子(epidermal growth factor receptor,EGFR)、蛋白激酶B(protein kinase B,AKT)、雷帕霉素靶蛋白(mammalian target of rapamycin,mTOR)的mRNA表达水平降低(P<0.05);EGFR、p-EGFR、AKT、p-AKT、mTOR蛋白表达水平降低(P<0.05)。结论ARG可抑制鼻咽癌5-8F细胞裸鼠移植瘤的生长,其机制可能与下调EGFR、p-EGFR、AKT、p-AKT、mTOR蛋白的表达有关。
OBJECTIVE To study the effect of arctigenin(ARG)on the growth of transplanted tumor of NPC 5-8F cells in nude mice and to explore the molecular mechanism.METHODS 5-8F cells of NPC were injected into the right armpit of nude mice to establish the transplanted tumor model of NPC 5-8F cells.The effect of ARG on the growth of transplanted tumor of NPC 5-8F cells was evaluated by intraperitoneal injection of drugs.Real-time PCR and Western blotting were used to detect the mRNA expression level and the protein expression level.RESULTS ARG could inhibit the growth of transplanted tumor of NPC 5-8F cells.Compared with the control group,ARG could significantly reduce the mRNA expression of EGFR,Akt and mTOR and down regulate the protein expression of EGFR,p-EGFR,AKT,p-AKT and mTOR(P<0.05).CONCLUSION ARG could inhibit the growth of transplanted tumor of NPC 5-8F cells in nude mice,and its mechanism might be related to down regulating the protein expression of EGFR,p-EGFR,AKT,p-AKT and mTOR.
