摘要
恶性肿瘤与慢性炎症有着密切关系,慢性炎症中促炎症细胞因子通过激活经典的IKKα和IKKβ途径,从而激活NF-кB途径,抑制细胞凋亡,加快细胞周期,促进肿瘤发生和肿瘤发展,其中IKKβ在经典的IKKα和IKKβ途径中起着主导作用。阻断IKKα和IKKβ途径,通过增加凋亡,减少促炎症因子的表达,缩小了肿瘤的体积。因此,阻断IKKα和IKKβ途径是治疗恶性肿瘤一个新的作用靶点。目前,已发现一些阻断IKKα和IKKβ途径的药物具有良好的抗肿瘤作用。
Malignant tumor is closely related to chronic inflammation. The proinflammation factors activate canonical IKKα and IKKβ thus activating the NF-кB signaling pathway, inhibiting the apoptosis of tumor cells, accelerating the cell cycle,and facilitating the tumorigenesis and tumor profession. IKKβ plays a central role in canonical IKKα and IKKβ pathway. The inhibition of IKKα and IKKβ markedly reduces the expression of proinflammation factors, augmented apoptosis, and decreases the volume of tumor. Therefore, IKKα and IKKβ pathways are novel targets for treating cancer. Some anti-tumor drugs have shown promising efficacy in treating tumors by suppressing these pathways.
