摘要
目的:探讨芍药提取物对高脂诱导的大鼠认知功能的影响及机制,为临床改善精神分裂症认知功能障碍提供新的思路和线索。方法:选取40只雄性SD大鼠,均给予高脂高糖饲料喂养,大鼠随机分为空白对照组、模型对照组、二甲双胍组、芍药提取物实验组各10只,分别灌胃生理盐水、0.15 g/kg二甲双胍溶液,5 mg/mL芍药提取物。给药结束后检测大鼠体质量,性腺脂肪量,血糖和糖化血红蛋白,Morris水迷宫系统测定各组大鼠认知功能,蛋白免疫印迹实验检测PI3K、AKT蛋白表达水平。结果:与模型对照组比较,二甲双胍组、芍药提取物实验组大鼠体质量,体质量,性腺脂肪量,血糖和糖化血红蛋白均降低(P<0.05);大鼠的逃避潜伏期、探索距离显著减少,目标象限游泳时间显著增长、站台穿越次数显著增加;大鼠海马组织中PI3K、phospho-AKT(S473)蛋白的表达水平降低。结论:芍药提取物可降低高脂诱导的大鼠的体质量和血糖水平,改善认知功能,其机制与抑制PI3K/AKT信号通路相关。芍药提取物可能成为治疗精神分裂症认知功能障碍的新方法。
Objective:To investigate the effect and mechanism of paeonia lactiflora extract on the cognitive function of high-fat diet induced rats so as to provide new ideas for clinical treatment of cognitive dysfunction in schizophrenia.Methods:A total of 40 male SD rats were feed with high-fat and high-glucose diet,and they were divided into a blank control group,a model control group,a metformin group,a paeonia lactiflora extract group with 10 cases in each group.The 10 rats were respectively orally given with normal saline,0.15 g/kg metformin solution,5 mg/mL paeonia lactiflora extract.After the administration,the rats′body weight,gonadal fat mass,blood glucose and glycosylated hemoglobin were measured.The Morris water maze system was used to measure the cognitive function of rats in each group,and the protein expression levels of PI3K and AKT were measured by Western blotting.Results:Compared with the model control group,the weight,body weight,gonadal fat mass,blood sugar and glycosylated hemoglobin of rats in the metformin group and the peony extract experimental group were reduced(P<0.05);the escape latency and exploration distance of rats were significantly reduced,and the target quadrant swimming time increased significantly and the number of platform crossings increased significantly;the expression levels of PI3K and phospho-AKT(S473)proteins in the hippocampus of rats decreased.Conclusion:Paeonia lactiflora extract can reduce the body weight and blood glucose levels in high-fat diet induced rats,and improve their cognitive function.The mechanism is related to the inhibition of the PI3K/AKT signaling pathway.Paeonia lactiflora extract may be a new way for the treatment of cognitive dysfunction in schizophrenia.
