摘要
目的研究宫颈病变与Treg转录因子表达及细胞因子水平的关系。方法选择2015年3月至2018年12月期间在本院接受手术切除的宫颈癌、宫颈上皮内瘤变(CIN)患者及子宫肌瘤患者,分别留取宫颈癌组织、CIN组织、正常宫颈组织,检测Foxp3及增殖基因cyclinD1、p53的表达、IL-10、TGF-β1的含量。结果宫颈癌组织中Foxp3、cyclinD1的表达及IL-10、TGF-β1的含量明显高于CIN组织和正常宫颈组织,p53的表达明显低于CIN组织和正常宫颈组织,差异有统计学意义(P<0.05);高危HPV感染、FIGO分期Ⅲ^Ⅳ期、低分化、有淋巴结转移的宫颈癌组织中Foxp3的表达及IL-10、TGF-β1的含量明显高于无高危HPV感染、FIGO分期Ⅰ^Ⅱ期、中高分化、无淋巴结转移的宫颈癌组织,差异有统计学意义(P<0.05);Foxp3的表达及IL-10、TGF-β1的含量与CyclinD1的表达呈正相关,与p53的表达呈负相关。结论 Treg转录因子Foxp3表达及细胞因子IL-10、TGF-β1含量增加与宫颈癌的发生与发展有关。
Objective To study the relationship between cervical lesions and Treg transcription factor expression and cytokine levels.Methods Patients with cervical cancer,cervical intraepithelial neoplasia(CIN)and uterine leiomyoma who underwent surgical resection in our hospital from March 2015 to December2018 were selected.Cervical cancer tissues,CIN tissues and normal cervical tissues were collected,the expression of Foxp3,cyclinD1,p53 and the contents of IL-10,TGF-β1 were detected.Results The expressions of Foxp3,cyclinD1 and the levels of IL-10,TGF-β1 in cervical cancer tissues were significantly higher than those in CIN tissues and normal cervical tissues.The expression of p53 in cervical cancer tissues were significantly lower than those in CIN tissues and normal cervical tissues.The difference was statistically significance(P<0.05).The expressions of Foxp3,cyclinD1 and the levels of IL-10,TGF-β1 in cervical cancer tissues with high-risk HPV infection,FIGO stageⅢ-Ⅳ,poor-differentiated,lymph node metastasis were significantly higher than those in cervical cancer tissues without high-risk HPV infection,FIGO stageⅠ-Ⅱ,moderately to well-differentiated,non-lymph node metastasis.The difference was statistically significance(P<0.05).The expression of Foxp3 and the levels of IL-10 and TGF-β1 were positively correlated with the expression of CyclinD1,and negatively correlated with the expression of p53.Conclusion The expression of Treg transcription factor Foxp3 and the increase of IL-10 and TGF-β1 are related to the occurrence and development of cervical cancer.
