血清4型禽腺病毒山东株基因组序列的克隆及遗传演化分析 被引量:2

摘要 从山东某地区发生心包积液综合征的发病鸡中收集病料,按常规方法处理后接种SPF鸡胚,分离到1株病毒,经PCR和测序分析鉴定该分离株为Ⅰ群禽腺病毒、血清4型禽腺病毒,命名为FAV-SD。合成引物成功扩增病毒全基因组序列,并进行序列的测定与分析,结果表明FAVSD株全基因组大小为43752 bp,通过hexon基因同源性比对,FAV-SD与我国今年报道的禽腺病毒血清4型同源性最高;遗传进化分析结果表明,FAV-SD分离株与我国禽腺病毒血清4型流行毒株处于同一分支,而与国外禽腺病毒血清4型毒株不在同一分支,存在遗传差异,说明中国流行的血清4型禽腺病毒具有自身地域特点。该病毒株的分离鉴定为禽腺病毒疫苗的研制奠定了基础。 Pathogenic specimens were collected from sick chickens with pericardial effusion syndrome from an area of Shandong.After the specimens were treated by the conventional method,they were inoculated with specific pathogen-free chicken embryos,and one strain of virus was isolated.After PCR and sequencing analysis,this isolate was identified as groupⅠserotype 4 fowl adenovirus and was named as fowl adenovirus(FAdV)-SD2016.The synthesized primers were able to successfully amplify the whole genome sequence of this virus,and its sequence was determined and analysed.The results showed that the size of the whole genome of the FAdV-SD2016 strain was 43,752 bp.By comparing the homology of the hexon gene,FAdV-SD2016 had the highest homology with the fowl adenovirus serotype 4,which was reported in China earlier this year.Genetic evolution analysis results showed that the FAdV-SD2016 isolate was in the same branch as the Chinese epidemic strain of the fowl adenovirus serotype 4,but it was not in the same branch as the foreign strain of the fowl adenovirus serotype 4.This genetic difference suggested that the fowl adenovirus serotype 4 that was prevalent in China had its own regional characteristics.The isolation and identification of this viral strain laid a foundation for the development of an fowl adenovirus vaccine.
