新型鸭呼肠孤病毒分离鉴定及σC基因进化分析

Isolation and phylogenetic analysis ofσC gene of a novel duck orthoreovirus

新型鸭呼肠孤病毒(Novel duck orthoreovirus,NDRV)在番鸭和北京鸭上流行,引起番鸭的肝斑状出血和北京鸭的脾坏死。本研究从广东云浮某养殖场发生以肝脏出血为病变的番鸭中分离到一株NDRV,命名为GD-YF-202001。对GD-YF-202001株的σC基因进行RT-PCR扩增、克隆测序与序列分析。结果显示,分离株σC基因与NDRV SDJN18毒株同源性最高,遗传距离较近;分离株σC氨基酸序出现多个氨基酸位点突变,发生了较大的变异。

The circulation of novel duck orthoreovirus(NDRV)in Muscovy ducks and Peking ducks causes hemorrhage in the liver of Muscovy ducks and necrosis in the spleen of Peking ducks.In this study,a NDRV strain GD-YF-202001 was isolated from Muscovy ducks exhibited liver hemorrhage in Yunfu city,Guangdong province.TheσC gene of GD-YF-202001 strain was amplified by RT-PCR.Sequence analysis results showed that theσC gene of the isolate had high identities and phylogenetic close with NDRV SDJN18 strain.The amino acid sequence of theσC protein of the isolate has a lot of residue mutation compared with the referent strain,which indicates that great variation has occurred in the isolate.
