淫羊藿苷对去势雌性高血压大鼠内皮祖细胞的类雌激素保护作用研究 被引量:6

摘要 目的探索淫羊藿苷对去势雌性高血压大鼠体重、血压、心率及内皮祖细胞功能的影响。方法9周龄去势雌性京都Wistar大鼠(Wistar Kyoto rats,WKY)10只,9周龄去势雌性自发高血压大鼠(spontaneously hypertensive rat,SHR)50只,以WKY大鼠为空白组,SHR组大鼠随机分为模型组、雌激素组、淫羊藿苷低剂量组、淫羊藿苷高剂量组、淫羊藿苷低剂量+雌激素受体抑制剂组,分别给予相应药物8周。自给药起每周检测各组大鼠体重,检测给药前及给药8周后各组大鼠平均动脉压及心率,并于给药8周后麻醉状态下腹主动脉取血提取外周血内皮祖细胞,处死大鼠后分离其后肢提取大鼠骨髓内皮祖细胞。体外鉴定并培养内皮祖细胞,并进行内皮祖细胞活性及NO分泌水平检测。结果(1)给药8周后,雌激素组大鼠体重与模型组相比明显下降(P<0.05);(2)给药前,与空白组比较,其他各组平均动脉压明显升高(P<0.05);给药8周后,模型组平均动脉压与空白组比较明显升高(P<0.05)。与模型组比较,雌激素组、淫羊藿苷低剂量及高剂量组平均动脉压明显下降(P<0.05);与淫羊藿低剂量组比较,淫羊藿低剂量+雌激素受体抑制剂组血压明显升高(P<0.05);(3)与给药前比较,给药8周后各组心率均明显升高(P<0.05);给药8周后,淫羊藿苷低剂量及高剂量组心率与模型组比较皆明显下降(P<0.05);(4)给药8周后,与空白组相比,模型组骨髓及外周血的内皮祖细胞活性及NO分泌能力明显下降(P<0.05);与模型组相比,雌激素组、淫羊藿苷低剂量及高剂量组均明显升高(P<0.05);与淫羊藿低剂量组比较,淫羊藿低剂量+雌激素受体抑制剂组明显下降(P<0.05)。结论淫羊藿苷作为一种植物类雌激素对去势雌性高血压大鼠血压、心率及内皮祖细胞具有保护作用。 Objective To explore the effect of icariin on body weight,blood pressure,heart rate and endothelial progenitor cells function in castrated female hypertension rats.Methods There were 10 female Wister Kyoto(WKY)castrated rats at 9 weeks old and 50 female Wister Kyoto(SHR)castrated rats at 9 weeks old.WKY rats were taken as the blank group,and SHR rats were randomly divided into 5 groups,including model group,estrogen group,low and high dose of icariin groups,and low dose of icariin combined with estrogen receptor antagonism group.Each group was given corresponding drug for 8 weeks,respectively.The body weight of the rats in each group was measured weekly.The mean arterial pressure and heart rate of the rats in each group were measured before and after 8 weeks of drug administration.The peripheral endothelial progenitor cells were extracted from the abdominal aorta under anesthesia 8 weeks after administration.After the rats were sacrificed,the rat bone marrow endothelial progenitor cells were isolated from their hind limbs.Endothelial progenitor cells were identified and cultured in vitro and their activity and nitric oxide(NO)secretion levels were detected.Results(1)After 8 weeks of administration,the body weight of rats in the estrogen group decreased significantly than in the model group(P<0.05);(2)Before administration,compared with blank group,the mean arterial pressure of other groups increased significantly(P<0.05);After 8 weeks of administration,the mean arterial pressure in model group increased significantly than that in blank group(P<0.05);Compared with model group,the mean arterial pressure in estrogen group,low and high dose of icariin groups decreased significantly(P<0.05);Compared with low dose of icariin group,the mean arterial pressure in low dose of icariin combined with estrogen receptor antagonism group increased significantly(P<0.05).(3)Compared with before administration,the heart rate in each group increased significantly after 8 weeks administration(P<0.05);After 8 weeks of administration,compared with the model group,the heart rate in low and high dose of icariin groups decreased significantly than that in model group(P<0.05).(4)After 8 weeks of administration,the activity and NO secretion of endothelial progenitor cells in bone marrow and peripheral decreased significantly than those in the model group(P<0.05);Compared with model group,those indexes in estrogen group,low and high dose of icariin groups increased significantly(P<0.05).Compared with low dose of icariin group,those indexes in low dose of icariin combined with estrogen receptor antagonism group decreased significantly(P<0.05).Conclusion Icariin,as an implant estrogen,has protective effects on blood pressure,heart rate and endothelial progenitor cells in castrated female hypertension rats.
