基于辨证保健理论探讨红曲茯苓片对脾虚湿盛型高脂血症大鼠的影响 被引量:9

摘要 目的观察红曲茯苓片对脾虚湿盛型高脂血症大鼠的影响。方法将SD大鼠分为空白组、复合模型组、血脂康阳性药组,以及红曲茯苓片低、中、高剂量组共6组,采用连续28天给予高脂饲料,及每日力竭游泳复合饮食不节(饱食1天,禁食1天,自由饮水)造脾虚湿盛型高脂血症模型,观察红曲茯苓片对于大鼠进食量、体重的影响,ELISA测定血清甘油三脂、总胆固醇、高密度脂蛋白、低密度脂蛋白、胃泌素、胃动素、胰蛋白酶、胆碱酯酶、白蛋白、醛固酮、血管活性肠肽的含量,免疫组化法测定胃、肠组织中水通道蛋白3、水通道蛋白4的含量。结果(1)与空白组相比,复合模型组进食量、体重、胃泌素、胃动素、胰蛋白酶、白蛋白含量显著降低(P<0.01),胃、肠组织中水通道蛋白3表达量降低(P<0.01),甘油三酯、总胆固醇、低密度脂蛋白、胆碱酯酶、醛固酮、血管活性肠肽水平升高(P<0.05、P<0.01),胃、肠组织中水通道蛋白4表达量升高(P<0.01)。(2)与复合模型组相比,红曲茯苓片和血脂康都能增加进食量、体重、高密度脂蛋白、胃泌素、胃动素、胰蛋白酶水平以及胃、肠组织中水通道蛋白3表达量(P<0.05、P<0.01),红曲茯苓片还能提高白蛋白水平(P<0.05)。红曲茯苓片和血脂康都能降低甘油三脂、总胆固醇、低密度脂蛋白、胆碱酯酶、血管活性肠肽水平以及胃、肠组织中水通道蛋白4表达量(P<0.05、P<0.01),红曲茯苓片还能降低醛固酮水平(P<0.05)。(3)与血脂康组相比,红曲茯苓片高剂量组能增加进食量、胃泌素、胃动素水平以及肠组织中水通道蛋白3、水通道蛋白4的表达量(P<0.01),降低低密度脂蛋白水平(P<0.01),红曲茯苓片低剂量组、中剂量组能提高总胆固醇水平以及肠组织中水通道蛋白3、水通道蛋白4的表达量(P<0.05、P<0.01),中剂量组能升高胃泌素水平(P<0.01),低剂量组能降低胰蛋白酶水平(P<0.05)。结论红曲茯苓片能降低脾虚湿盛型高脂血症大鼠血脂水平,具有健脾除湿功效。 Objective To observe effects of Hongqu Fuling tablets on hyperlipidemia rats with overabundant dampness due to spleen deficiency.Methods SD rats were divided into 6 groups,including blank group,compound model group,Xuezhikang positive drug group,low,medium and high doses of Hongqu Fuling tablets groups.The hyperlipidemia model with overabundant dampness due to spleen deficiency was established by giving high-fat feed and daily exhausted swimming combined diet(one day satiety,one day fasting,and free drinking water)for 28 consecutive days.The effects of Hongqu Fuling tablets on the intake and body weight of rats were observed.The contents of serum triglycerides,total cholesterol,high-density lipoprotein,low-density lipoprotein,gastrin,motilin,trypsin,cholinesterase,albumin,aldosterone and vasoactive intestinal peptide were determined by enzyme linked immunosorbent assay.The contents of aquaporin-3 and aquaporin-4 in gastric and intestinal tissues were determined by immunohistochemistry.Results(1)Compared with the blank group,in the compound model group,the contents of food intake,body weight,stomach secrete hormone,dynamic element,trypsin and albumin were decreased significantly(P<0.01),the expression levels of aquaporin-3 in the stomach and intestinal tissue were decreased(P<0.01),the levels of triglycerides,total cholesterol,low density lipoprotein,cholinesterase,aldosterone and vasoactive intestinal peptide were increased(P<0.05,P<0.01),the expression levels of aquaporin-4 in the stomach and intestinal tissue were increased(P<0.01).(2)Compared with the compound model group,both Hongqu Fuling tablets and Xuezhikang could increase the amount of food intake,body weight,high-density lipoprotein,gastrin,motility factor,trypsin and the expression of aquaporin-3 in the stomach and intestinal tissues(P<0.05,P<0.01),and Hongqu Fuling tablets could also increase the albumin level(P<0.05).(3)Compared with the Xuezhikang group,high dose of Hongqu Fuling tablets group could increase the levels of food intake,stomach secrete hormone,motilin,aquaporin-3 and aquaporin-4 in the stomach and intestinal tissues(P<0.01),lowering the level of density lipoprotein(P<0.01),low and medium dose of Hongqu Fuling tablets groups could increase the level of total cholesterol,aquaporin-3 and aquaporin-4 in the stomach and intestinal tissues(P<0.05,P<0.01),medium dose of Hongqu Fuling tablets group could increase the level of stomach secrete hormone(P<0.01),low dose of Hongqu Fuling tablets group could decrease trypsin(P<0.05).Conclusion Hongqu Fuling tablets can reduce the blood lipid level of hyperlipidemia rats with overabundant dampness due to spleen deficiency,and it has the effect of invigorating spleen for eliminating dampness.
