摘要
目的检测天冬酰胺连接糖基化3(ALG3)在人膀胱癌组织中的表达情况,分析其临床意义。方法采用生物信息学的方法分析ALG3在膀胱癌组织及正常组织中的mRNA水平,并分析其表达与膀胱癌患者的生存率间的关系。回顾性分析87例接受手术治疗的膀胱癌患者的临床病理资料。采用免疫组化法检测膀胱癌组织和癌旁健康组织中的ALG3蛋白表达水平,分析其与膀胱癌患者临床病理特征的关系。结果生物信息学分析结果显示,ALG3的mRNA在膀胱癌组织中显著高表达,并与患者总生存率和无病生存率显著相关。免疫组化结果表明,ALG3主要定位于胞浆,且在膀胱癌组织中高表达,而在癌旁健康组织中低表达或无表达。ALG3在膀胱癌肿瘤组织中的高表达与肿瘤分期和肿瘤复发相关(均P<0.05),而与年龄、性别、肿瘤分级和淋巴结转移无关(均P>0.05)。结论ALG3在膀胱癌发生和发展中发挥重要作用,其有望成为膀胱癌重要的治疗靶点和预后标志物。
Objective To detect the expression of ALG3 in human bladder cancer and analyze its clinical significance.Methods Bioinformatics was conducted to analyze the mRNA level of ALG3 in bladder cancer tissues and normal tissues,and to analyze the relationship between its expression and the survival rate of bladder cancer patients.The clinical and pathological data of 87 patients with bladder cancer who underwent surgical treatment were retrospectively analyzed.Immunohistochemical method was used to detect the expression level of ALG3 protein in bladder cancer tissues and healthy tissues adjacent to the cancer,and to analyze its relationship with clinicopathological characteristics of bladder cancer patients.Results The results of bioinformatics analysis showed that ALG3 mRNA was highly expressed in bladder cancer tissues,and was significantly correlated with the overall survival rate and disease-free survival rate of patients.Immunohistochemical results showed that ALG3 was mainly localized in the cytoplasm and was highly expressed in bladder cancer tissues,but low or no expression in healthy tissues adjacent to the cancer.The high expression of ALG3 in bladder cancer tumor tissue was related to tumor stage and tumor recurrence(all P<0.05),but not related to age,gender,tumor grade and lymph node metastasis(all P>0.05).Conclusions ALG3 plays an important role in the occurrence and development of bladder cancer,and it is expected to become an important therapeutic target and prognostic marker for bladder cancer.
