黄连抗胃溃疡炎症的作用机制及其活性成分的实验研究 被引量:1

Experimental study on the mechanism of action of Coptis chinensis Franch.against gastric ulcer inflammation and its active components

摘要 目的以大鼠胃溃疡为炎症模型,黄连为给予药物,研究其抗胃溃疡炎证的药效与化学成分之间的谱效关系,明确其抗胃溃疡炎症的活性成分。方法建立12批不同来源的黄连HPLC指纹图谱,并进行药效学实验;用灰色关联分析方法对指纹图谱16个共有峰的峰面积和抗炎作用的相关性进行分析。结果 16个共有峰与血清白细胞介素-3(IL-3)的关联度均>0.6,与血清中肿瘤坏死因子-α的关联度除1号峰以外均>0.6,说明各峰对黄连抗胃溃疡炎症作用均具相关性,其中已知成分盐酸药根碱、盐酸小檗碱、盐酸巴马汀抗炎活性大于其他已知成分的活性,未知成分3、5、8、15号峰抗炎活性较其他未知成分抗胃溃疡炎症活性大。结论通过黄连抗胃溃疡实验和谱效关联分析研究,结果表明黄连抗胃溃疡炎症是多成分共同作用的结果。 Objective Rat gastric ulcer was used as an inflammatory model, and Coptis chinensis was used as a therapeutic drug to study the spectral effect relationship between its anti-gastric ulcer syndrome and chemical composition, and to identify its active ingredients against gastric ulcer inflammation. Methods The HPLC fingerprints of 12 batches of Coptis chinensis from different sources were established and pharmacodynamic experiments were carried out. The correlation between the peak area of 16 common peaks of fingerprints and anti-inflammatory efficacy was analyzed by grey correlation analysis. Results The correlation between 16 common peaks and serum interleukin-3(IL-3) was >0.6, and the correlation with serum tumor necrosis factor-α was >0.6 except for peak No.1, indicating that each peak is resistant to gastric ulcer inflammation. The effects are all related. The anti-inflammatory activity of known components such as jatrorrhizine hydrochloride, berberine hydrochloride and palmatine hydrochloride is greater than that of other known components. The anti-inflammatory activity of unknown components 3, 5, 8, and 15 is higher. Other unknown components have high inflammatory activity against gastric ulcer. Conclusion Through the anti-gastric ulcer test and spectral effect correlation analysis of Coptis chinensis, the results showed that Coptis chinensis anti-gastric ulcer inflammation is the result of multi-component synergy.
