重组脑利钠肽对家兔冠脉搭桥术静脉桥内皮功能和炎症水平的影响 被引量:1

Effects of recombinant brain natriuretic peptide on endothelial function and inflammatory level of vein graft in rabbits undergoing coronary artery bypass grafting

摘要 目的:探讨重组脑利钠肽对家兔冠脉搭桥术静脉桥内皮功能和炎性因子的影响。方法:30只家兔建立冠脉搭桥术,并随机分为模型组、低剂量组和高剂量组,低剂量组和高剂量组将耳缘静脉注射0.1 mg/(kg·d)和0.3 mg/(kg·d),模型组注射等体积生理盐水。同时选取10只家兔作为对照组,注射等体积生理盐水。4组家兔治疗6个月后,观察静脉桥狭窄程度、炎性因子和内皮功能。结果:与模型组(40.27±7.90)%比较,低剂量组和高剂量组家兔静脉桥血管狭窄程度(34.12±5.09)%和(26.74±4.88)%显著下降,差异有统计学意义(t=3.501、3.901,P<0.05)。与低剂量组比较,高剂量组家兔静脉桥血管狭窄程度显著下降,差异有统计学意义(t=2.817,P<0.05)。与模型组[(298.44±15.39)、(326.65±17.33)、(154.32±18.64)pg/ml]比较,低剂量组静脉桥炎性因子水平肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-1β和IL-6水平[(230.29±12.92)、(285.60±15.20)、(119.32±12.67)pg/ml]显著下降,差异有统计学意义(t=4.921、4.282、4.389,P<0.05)。与低剂量组比较,高剂量组家兔静脉桥炎性因子TNF-α、IL-1β和IL-6水平[(104.44±10.78)、(230.32±12.66)、(95.24±10.09)pg/ml]显著下降,差异有统计学意义(t=2.910、2.714、2.391,P<0.05)。与模型组[(120.39±19.48)μmol/ml、(12.65±1.07)ng/ml、(37.90±4.19)pg/ml]比较,低剂量组静脉桥内皮功能因子水平ET-1和vWF[(103.47±14.89)μmol/ml、(10.39±1.20)ng/ml]显著下降,NO水平[(51.36±6.19)pg/ml]显著增加,差异有统计学意义(t=3.594、3.709、3.100,P<0.05)。与低剂量组比较,高剂量组家兔静脉桥内皮功能因子ET-1和vWF[(87.29±9.03)μmol/ml、(230.32±12.66)ng/ml],NO水平显著增加[(64.22±5.27)pg/ml],差异有统计学意义(t=2.418、2.115、2.003,P<0.05)。结论:重组脑利钠肽可显著改善动脉搭桥术后静脉桥内皮功能和炎性因子水平,对静脉桥具有保护作用。 Objective To investigate the effects of recombinant human brain natriuretic peptide(rhBNP)on endothelial function and inflammatory factors in rabbits undergoing coronary artery bypass grafting.Methods 30 rabbits were randomly divided into model group,low-dose group and high-dose group.In low-dose group and high-dose group,0.1 mg/(kg·d)and 0.3 mg/(kg·d)were injected into ear vein,and the same volume of normal saline was injected into the model group.10 rabbits were selected as control group and injected with equal volume of normal saline.After 6 months of treatment,the degree of vein bridge stenosis,inflammatory factors and endothelial function were observed.Results Compared with the model group(40.27±7.90)%,the stenosis degree of vein bridge in low dose group and high dose group(34.12±5.09)%and(26.74±4.88)%(t=3.501,3.901,P<0.05).Compared with the low-dose group,the stenosis degree of the high-dose group significantly decreased(t=2.817,P<0.05).Compared with the model group[(298.44±15.39),(326.65±17.33),(154.32±18.64)pg/ml],the levels of tumor necrosis factor-α(TNF-α),interleukin(IL)-1βand IL-6 in the low-dose group[(230.29±12.92),(285.60±15.20),(119.32±12.67)pg/ml]significantly decreased in the low-dose group(t=3.719,3.518,3.209,P<0.05).Compared with the low-dose group,the levels of TNF-α,IL-1βand IL-6 in the high-dose group[(104.44±10.78),(230.32±12.66),(95.24±10.09)pg/ml]significantly decreased in the high-dose group(t=2.910,2.714,2.391,P<0.05).Compared with the model group[(120.39±19.48)μmol/ml,(12.65±1.07)ng/ml,(37.90±4.19)pg/ml],the levels of ET-1 and vWF[(103.47±14.89)μmol/ml,(10.39±1.20)ng/ml]in the low-dose group significantly decreased,and the level of NO[(51.36±6.19)pg/ml]significantly increased(t=3.594,3.709,3.100,P<0.05).Compared with the low-dose group,the levels of ET-1 and vWF[(87.29±9.03)μmol/ml,(230.32±12.66)ng/ml]in the high-dose group significantly reduced and the level of NO in high dose group[(64.22±5.27)pg/ml]significantly increased(t=2.418,2.115,2.003,P<0.05).Conclusion Recombinant brain natriuretic peptide can significantly improve endothelial function and inflammatory factor level of venous bridge after arterial bypass grafting,which has protective effect on venous bridge.
