摘要
目的探讨参苓白术散对支气管哮喘小鼠气道炎症的抑制作用及其对肠道菌群结构的调节作用。方法将SPF级雄性小鼠随机分为正常组、模型组、参苓白术散组、西药组,每组10只。通过抗生素诱导、致敏和雾化激发建立肠道菌群紊乱的哮喘小鼠模型。观察小鼠一般情况、肺组织病理变化,对各组小鼠粪便进行16S rRNA测序并分析治疗后肠道菌群的变化。结果参苓白术散组、西药组哮喘小鼠的生理状态、哮喘症状较模型组均好转,肺组织炎症低于模型对照组(P<0.05)。16S rRNA测序结果显示模型组小鼠肠道菌群的丰富度和多样性与正常组比均减少;参苓白术散治疗后小鼠肠道菌群的丰富度和多样性有所增加,且在优势菌群中,拟普雷沃菌属相对丰度增加明显。结论肠道菌群丰富度的改变及菌群结构的紊乱是哮喘发病的危险因素之一。参苓白术散改善哮喘气道炎症的作用可能与调节哮喘小鼠的肠道菌群有关。
Objective To investigate the inhibitory effect of Shen Ling Baizhu powder on airway inflammation in asthmatic mice and its regulatory effect on the structure of gut microbiota.Methods SPF grade male mice were randomly divided into normal group,model control group,Shen Ling Baizhu powder group and Western medicine group.The model of asthmatic mice with disturbed intestinal gut microbiota was established by antibiotic induction,sensitization and atomization.The general situation of mice,pathological changes of lung tissue were observed,and 16S rRNA of feces of mice in each group were sequenced and the changes of gut microbiota after treatment were analyzed.Results①The physiological status and asthma symptoms of asthmatic mice in Shen Ling Baizhu powder group and Western medicine group were better than those of control group.②Mice in Shen Ling Baizhu powder group and Western medicine group showed lower inflammation of lung tissue than those in model control group(P<0.05).③16S rRNA sequencing showed that the abundance and diversity of gut microbiota in model control group mice decreased compared with those in normal group.The richness and diversity of gut microbiota in mice were increased after treatment with Shen Ling Baizhu powder,and the relative abundance of the genus Prevo was significantly increased in the dominant flora.Conclusion The changes of gut microbiota richness and the disorder of flora structure are both the risky factors of asthma.The effect of Shen Ling Baizhu powder on airway inflammation may be related to regulating gut microbiota in asthmatic mice.
