摘要
目的建立泽泻药材高效液相色谱(HPLC)指纹图谱。方法采用Agilent ZORBAX Extend-C18色谱柱(250 mm×4.6 mm,5μm);以乙腈-水为流动相,梯度洗脱;流速:1.0 mL·min^-1;柱温:30℃;检测波长:208 nm;进样量:10μL。采用指纹图谱相似度评价软件,结合聚类分析(HCA)、主成分分析(PCA)和偏最小二乘法判别分析(PLS-DA)对24批泽泻药材指纹图谱进行评价。结果24批泽泻药材HPLC指纹图谱中有16个共有峰,鉴定出其中5个,分别为泽泻醇A、泽泻醇B、泽泻醇F、23-乙酰泽泻醇B、环氧泽泻烯。泽泻药材HPLC指纹图谱相似度在0.788~0.985之间,不同产地样品间质量存在较大差异。通过HCA、PCA、PLS-DA分析,将24批样品分为2类(建泽泻和江泽泻聚为1类,川泽泻聚为1类),并筛选出泽泻醇A、泽泻醇B、泽泻醇F、环氧泽泻烯以及未知成分峰7、峰9和峰10等7个差异性标志成分。结论该方法简便、快速、重复性好,可为泽泻药材的整体质量评价提供依据。
Objective To establish the HPLC fingerprint for Alismatis rhizoma.Methods The separation was performed on Agilent ZORBAX Extend-C18 column(250 mm×4.6 mm,5μm)with the mobile phase consisting of acetonitrile and water at a flow rate of 1.0 mL·min^-1.The column temperature was 30℃,the detection wavelength was at 208 nm,and the sample injection volume was 10μL.Similarity evaluation combined with hierarchical clustering analysis(HCA),principal components analysis(PCA)and discriminant analysis of partial least squares(PLS-DA)were used to evaluate the HPLC chromatographic fingerprint of 24 batches of Alismatis rhizoma.Results There were 16 common peaks in the HPLC chromatographic fingerprint of 24 batches of Alismatis rhizome,and 5 of them were identified as alisol A,alisol B,alisol F,alisol B 23-acetate,alismoxide,respectively.The similar degrees of 24 batches of Alismatis rhizoma were between 0.788 and 0.985,the quality of Alismatis rhizoma from different regions varies greatly.Twenty four batches of samples were classified into 2 clusters,with products of Fujian province and Jiangxi province were clustered into one group,and those of Sichuan being the other group.Seven markers were selected as alisol A,alisol B,alisol F,alismoxide and three unknown components.Conclusion This method is simple,fast,and repeatable and can be used for the quality control of Alismatis rhizoma.
