高效液相色谱法同时测定玳玳花中5个成分的含量

Simultaneous Determination of Five Components in Citrus aurantium L.var.amara Engl.by HPLC

目的建立高效液相色谱(HPLC)法同时测定玳玳花中芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、桔皮素的含量,为其质量控制提供参考。方法采用Shim-Pack GIST C18色谱柱(150 mm×4.6 mm,5.0μm);以0.1%磷酸水(A)-乙腈(B)为流动相,梯度洗脱;流速:1.0 mL·min-1;检测波长:283 nm。结果玳玳花中5个成分在考察的浓度范围内,浓度与峰面积之间呈良好的线性关系(r>0.999);回收率均在100.13%~103.98%范围内,RSD均小于3.0%。聚类分析可直观地把4个不同产地的样品进行分类。结论该方法简便、准确,可用于比较不同来源玳玳花的差异,并为其质量标准建立提供依据。

Objective To establish a UHPLC method for simultaneous determination of five components(Narirutin,Naringin,Hesperidin,Neohesperidin and Tangeretin)in Citrus aurantium L.var.amara Engl.,in order to provide evidences for its quality control.Methods The separation was performed on a Shim-Pack GIST C18 column(150 mm×4.6 mm,5.0μm)with gradient elution using 0.1%phosphoric acid(A)and acetonitrile(B)as mobile phase at a flow rate of 1.0 mL·min-1.The detection wavelength was 283 nm.Results All calibration curves were linear(r>0.999)over the tested ranges.The average recoveries ranged from 100.13%to 103.98%with RSD values below 3.0%.Statistics software can intuitively classify samples from four different origins.Conclusion The established method is simple,fast and with high precision,which can be used to compare the differences of different sources and provide references for establishment of quality standard for Citrus aurantium L.var.amara Engl.
