摘要
目的探讨长链非编码RNA CCAT1表达对人胶质瘤细胞U251增殖、侵袭能力的影响及其作用机制。方法选取人胶质瘤细胞U251进行转染实验,分为空白组、sh-NC组和sh-CCAT1组;利用实时荧光定量qRT-PCR检测抑制效果;采用MMT、Transwell试验检测下调CCAT1对于胶质瘤细胞增殖、侵袭能力的影响;利用Western blot检测丝裂原活化蛋白激酶(MAPK)信号通路相关分子的表达变化。结果在转染48h后,sh-CCAT1组CCAT1的表达较NC组和sh-NC组显著降低(P<0.05);CCAT1下调后,胶质瘤细胞U251的增殖、侵袭能力显著下降(P<0.05),蛋白表达检测表明,CCAT1下调显著抑制MAPK信号通路分子的表达。结论CCAT1可能通过MAPK通路抑制胶质瘤的增殖和侵袭。
Objective To investigate the expression of long noncoding RNA(lncRNA)CCAT1in glioma cells U251,and its role in proliferation and invasion.Methods Human glioma cells U251were transfected by shRNA and were divided into three groups:blank group,sh-NC group and sh-CCAT1group.Real-time quantitative PCR was used to detect the inhibition effect.The effects of down-regulation of CCAT1on the proliferation and invasion of glioma cells were detected by MTT and Transwell tests.Western blot was used to detect the expression changes of related molecules in the mitogen-activated protein kinase(MAPK)signaling pathway.Results After transfection,the expression of CCAT1in sh-CCAT1group was significantly lower than that in NC group and sh-NC group(P<0.05);After the downregulation of CCAT1,the proliferation and invasion ability of glioma cells U251significantly decreased(P<0.05),the protein expression detection showed that the down-regulation of CCAT1significantly inhibited the molecular expression of MAPK signaling pathway.Conclusion CCAT1 may inhibit the proliferation and invasion of glioma through the MAPK signaling pathway.
