壮督驱寒合剂对成骨细胞炎性因子轴miR-29a/TNF-α/β-catenin的影响

Effect of Zhuangdu Quhan Mixture(壮督驱寒合剂)on the Inflammatory Factor Axis miR-29a/TNF-α/β-Catenin of Osteoblasts

目的:探究壮督驱寒合剂对成骨细胞炎性因子轴miR-29a/TNF-α/β-catenin的影响。方法:成骨细胞hFOB1.19分为空白对照组、正常血清组、AS血清组、含药血清组。AS血清组的培养基中加入20%患者血清,含药血清组培养基中加入20%壮督驱寒合剂含药血清,共培养72 h。检测实验末各组细胞miR-29a/TNF-α/β-catenin的表达水平。结果:与空白对照组比较,AS血清组miR-29a/TNF-α/β-catenin的表达明显上调,差异有统计学意义(P<0.05)。含药血清组miR-29a/TNF-α/β-catenin的表达明显下调,差异有统计学意义(P<0.05)。结论:壮督驱寒合剂能下调miR-29a/TNF-α/β-catenin的表达从而抑制炎性因子释放,这可能是其治疗AS的重要机制之一。

Objective:To explore the effect of Zhuangdu Quhan Mixture on the osteoblast inflammatory factor axis miR-29 a/TNF-α/β-catenin.Methods:Osteoblasts hFOB1.19 were divided into blank control group,normal serum group,AS serum group,and drug-containing serum group.20%patient serum was added to the culture medium of the AS serum group,and 20%Zhuangdu Quhan Mixture-containing serum was added to the medium of the drug-containing serum group,and the culture was for 72 hours.The expression level of miR-29 a/TNF-α/β-catenin in each group was detected at the end of the experiment.Results:Compared with the blank control group,the expression of miR-29 a/TNF-α/β-catenin in the AS serum group was significantly up-regulated,and the difference was statistically significant(P<0.05).The expression of miR-29 a/TNF-α/β-catenin in the drug-containing serum group was significantly down-regulated,and the difference was statistically significant(P<0.05).Conclusion:Zhuangdu Quhan Mixture can down-regulate the expression of miR-29 a/TNF-α/β-catenin and inhibit the release of inflammatory factors,which may be one of its important mechanisms for the treatment of AS.
