林烟草(Nicotiana sylvestris)K+通道蛋白基因NsAKT 1克隆及生物信息学分析 被引量:1

摘要 AKT 1是一种重要的钾离子通道蛋白,负责植物体中K+的吸收和运输。为探讨烟草中AKT 1编码基因的序列结构和蛋白功能,以林烟草(N.sylvestris)为材料,以cDNA为模板对NsAKT 1基因进行同源克隆,同时对NsAKT 1编码蛋白的结构及功能进行预测。结果表明,NsAKT 1基因的开放阅读框全长2682 bp,可编码893个AA,分子量为100.375 kDa。利用ProtParam、SignalP 4.0软件及Tmpred等软件对NsAKT 1蛋白的结构预测显示,该蛋白是一个亲水性的酸性跨膜蛋白,无信号肽,为非分泌型蛋白。NsAKT 1主要定位于内质网膜、高尔基体、质膜等部位,其二级结构由48个α螺旋(α-helix)、35个延伸链(extended strand)、36个β转角(β-turn)及59个无规则卷曲(random coil)组成,三者间通过无规则卷曲(random coil)进行连接。本研究为进一步探讨NsAKT 1的生物学功能提供了理论依据。 The AKT 1 is an important potassium channel protein responsible for the absorption and transport of K+in plants.In order to investigate the sequence structure and protein function of AKT 1 encoding gene in tobacco,this paper homologous cloned NsAKT 1 gene with N.sylvestris as the material and cDNA as the template,and predicted the structure and function of the protein encoding NsAKT 1.The results showed that the full length of the open reading frame of NsAKT 1 gene was 2682 bp,encoding 893 AA,and the molecular weight was 100.375 kDa.The structure prediction of N sAKT 1 protein by ProtParam,SignalP 4.0,Tmpred and other software showed that the protein was a hydrophilic acid transmembrane protein with no signal peptide and was a non-secreted protein.NsAKT 1 was mainly located in the endoplasmic reticulum omentum,golgi body and plasma membrane,etc.Its secondary structure was composed of 48 ben-helix,35 extended strand,36 bento-turn and 59 random coil,which were connected by random coil.This study provides a theoretical basis for further exploring the biological functions of NsAKT 1.
