基于网络药理学探讨黄芩-茵陈药对治疗儿童肺炎支原体肺炎的作用机制 被引量:4

摘要 目的:基于网络药理学方法探讨黄芩-茵陈药对治疗儿童肺炎支原体肺炎(MPP)的关键靶点及作用机制。方法:通过网络药理学方法,从药物和疾病两方面构建网络。从TCMSP数据库检索黄芩、茵陈的主要活性成分;DrugBank数据库、Swiss Target Prediction数据库查找并预测成分相应靶点;OMIM和GeneCards数据库收集MPP疾病靶点。整合药物靶点与疾病靶点,明确黄芩-茵陈药对治疗MPP的关键靶点及信号通路。结果:最终确定黄芩-茵陈药对的有效活性成分49个,预测潜在药物靶点123个;与MPP共有靶点27个;GO功能富集分析结果显示,该药对作用于本病的主要生物学功能涉及细胞因子活性、细胞周期调控、发热的正调节、平滑肌细胞增殖的正调控、趋化因子生物合成过程的正调控、DNA结合转录因子活性的调控、氧化应激反应等;与肿瘤坏死因子(TNF)、缺氧诱导因子(HIF-1)、T细胞受体、类节点受体、Toll样受体、丝裂原活化蛋白激酶(MAPK)、血管内皮生长因子(VEGA)等信号通路相关。结论:黄芩-茵陈药对治疗MPP具有多成分、多靶点、多途径的特点。主要干预炎症反应、病原微生物、免疫调节,从调控细胞周期与调节炎症免疫两方面发挥作用。 Objective:To explore the key target and mechanism of Huangqin(Scutellariae Radix)-Yinchen(Artemisiae Scopariae Herba)herbal pair in the treatment of mycoplasma pneumoniae pneumonia in children based on network pharmacology method.Method:The network was constructed from the drug and disease two aspects through the network pharmacology method.The main active components of Huangqin-Yinchen herbal pair were retrieved from TCMSP database.The DrugBank database and Swiss Target Prediction database were searched to predict the corresponding target of components.OMIM and GeneCards databases were searched to collect the disease targets.By integrating drug targets and disease targets,the key targets and signaling pathways of Huangqin-Yinchen herbal pair in the treatment of mycoplasma pneumoniae pneumonia in children were identified.Results:A total of 49 effective active ingredients of Huangqin-Yinchen herbal pair were finally identified,and 123 potential drug targets were predicted.There were 27 targets in total with mycoplasma pneumoniae pneumonia in children.GO function enrichment analysis showed the main biological functions of Huangqin-Yinchen herbal pair involved cytokine activity,cell cycle regulation,positive regulation of fever,positive regulation of smooth muscle cell proliferation,positive regulation of chemokine biosynthesis,regulation of DNA-binding transcription factor activity,oxidative stress reaction,etc..It is associated with tumor necrosis factor(TNF),hypoxia inducible factor(HIF-1),T cell receptor,node-like receptor,Toll-like receptor,mitogen-activated protein kinase(MAPK),vascular endothelial growth factor(VEGA)and other signaling pathways.Conclusions:Huangqin-Yinchen herbal pair has the characteristics of multi-component,multi-target and multi-pathway in the treatment of MPP.It mainly interferes with inflammatory response,pathogenic microorganisms and immune regulation,and plays a role in regulating cell cycle and inflammatory immunity.
