摘要
为明确鹅圆环病毒(Goose circovirus,GoCV)ORF3基因特征,本研究通过对朗德鹅源GoCV代表株(JX1株)基因组分析发现,JX1株ORF3基因全长为300 bp,编码99个氨基酸,未发现典型的核定位信号。所编码ORF3蛋白理论等电点为8.84、不稳定指数为80.95,为不稳定蛋白;脂肪系数为89.70;总平均疏水性指数为-0.472。ORF3基因核苷酸同源性分析比较发现,JX1株与yk1株(AY633653)、yk3株(DQ192280)核苷酸同源性最高,均为100%;与DG1株(KT808650)的核苷酸同源性最低,仅为89.0%。ORF3基因遗传进化分析发现,ORF3基因的进化频率并不完全与GoCV全基因组一致,ORF3基因在遗传进化上表现出明显的地域性,但不表现出时间性。本研究首次对GoCV ORF3基因特征进行分析探讨,为后续开展GoCV ORF3基因生物学功能研究奠定基础。
In order to identify the ORF3 gene characterization of goose circovirus(GoCV),in this study,Landes goose origin GoCV(JX1 strain)genome was used and analyzed.The results showed the ORF3 gene of JX1 strain was 300 nucleotides in length,coding 99 amino acids,which no typical nuclear localization signal(NLS)was found.The coding ORF3 protein theoretical pI,instability index,aliphatic index and grand average of hydropathicity was 8.84,80.95(means unstable protein),89.70 and-0.472,respectively.Nucleotides homology analysis shares a highly identity of 100%with yk1 strain(AY633653)、yk3 strain(DQ192280),but only 89.0%with DG1 strain(KT808650).Phylogenetic evolution analysis revealed that the evolutionary frequency of ORF3 gene did not math with full-genome,which showed typical regional but not time characteristic.In conclusion,this study firstly showed the genetic characterization the ORF3 gene of GoCV,which lays a good foundation for further genetic function research.
