苜蓿细胞周期蛋白CYCD5;1基因表达及其调控秋眠性机理研究

摘要 为探索苜蓿细胞周期蛋白CycD5;1基因在秋眠型紫花苜蓿(Medicago sativa L.)中表达调控规律及其调控苜蓿秋眠性的机理研究,通过检测春、夏、秋季和人工条件下不同温度、不同日照长度下秋眠型苜蓿Maverick叶片中CycD5;1基因mRNA相对表达量,分析其不同时期的差异,及其变化与株高、叶面积、日照长度、温度的相关性。结果表明:CycD5;1基因的表达受日照长度极显著正调控(P<0.01),受温度极显著负调控(P<0.01),但温度对其表达的调控占绝对优势;秋眠型苜蓿叶中CycD5;1基因mRNA相对表达量与株高、叶面积呈显著负相关关系(P<0.05)。研究表明,从夏季到秋季随着温度的降低,CycD5;1基因高表达很可能促进秋眠型苜蓿的秋眠。 To explore the expression regulation of CycD5;1 gene in alfalfa(Medicago sativa L.)leaves and its role in alfalfa fall dormancy,the study detected the mRNA relative abundance of CycD5;1 in leaves of fall dormant alfalfa Maverick in spring,summer and autumn,and at different temperature or different day lengths under artificial conditions.Differences among CycD5;1 mRNA relative abundance at different growth period were analyzed.The correlation among CycD5;1 mRNA relative abundance,plant height,leaf area,day length and temperature was analyzed.The results showed that CycD5;1 gene expression was very significantly negatively regulated by temperature(P<0.01)and highly significantly positively regulated by day length(P<0.01),and regulation of CycD5;1 gene expression in leaves of Maverick alfalfa was dominated by temperature;CycD5;1 mRNA relative abundance in leaves of Maverick alfalfa was significantly negatively correlated with plant height and leaf area(P<0.05).It was speculated that the high-expression of CycD5;1 promoted the fall dormancy of alfalfa with the decrease of temperature.
