茯砖茶中冠突散囊菌的分离及培养条件优化 被引量:6

Isolation and Optimization of Culture Conditions of Eurotium Cristatum from Fuzhuan Tea

摘要 为探究冠突散囊菌(Eurotium cristatum)的培养条件,以茯砖茶为原料,对该菌进行分离、纯化、鉴定,确定最佳培养基。在单因素实验基础上,采用L9(3^3)正交实验设计,对影响冠突散囊菌生长的匀浆时间、初始pH、接种量3个因素进行优化。结果表明:该菌孢子形态和单点式培养所得菌株具有冠突散囊菌的典型特征,马铃薯葡萄糖琼脂培养基是最适培养基,最佳培养条件为匀浆时间60 s、初始pH5.0、接种量0.4%。该研究可为冠突散囊菌的开发利用提供参考。 In order to explore the best culture conditions of Eurotium cristatum from Fuzhuan tea,Eurotium cristatum was isolated,purified,identified and the optimum culture medium was determined in this experiment.The factors including homogenization time,initial pH and inoculum size were optimized through single factor experiment and L9(3^3)orthogonal design.The results showed that the spore morphology of the fungus and the strains obtained by single point culture had the typical characteristics of Eurotium cristatum.The potato dextrose agar was the most suitable for the growth of Eurotium cristatum.The optimal culture conditions were homogenization time 60 s,initial pH 5.0 and inoculum size 0.4%,respectively.The study provided a theoretical basis for the development and utilization of Eurotium cristatum.
