铁死亡诱导剂和抑制剂的研究进展

Research progress in inducers and inhibitors of ferroptosis

铁死亡是一种新型细胞死亡方式,具有铁依赖性,表现为铁超载、脂质过氧化物蓄积和线粒体膜密度增加等特点,与癌症、心肌梗死、缺血性卒中和神经退行性疾病等生理病理过程密切相关。目前已发现多种铁死亡诱导剂或抑制剂,它们可通过不同的作用靶点和机制诱导或抑制铁死亡的发生,在治疗上述多种疾病方面具有潜在的临床价值。本文从结构、作用靶点、作用特点和应用模型等方面分类综述了铁死亡诱导剂和抑制剂的研究进展,为其进一步研究和临床应用提供理论参考。

Ferroptosis is a novel form of cell death in an iron-dependent manner,which is charac⁃terized by iron overload,lipid peroxides accumulation and condensed mitochondrial membrane densi⁃ties.Ferroptosis is closely related to the physiological and pathological processes such as cancer,myocardial infarction,ischemic stroke and neurodegenerative diseases.A variety of ferroptosis inducers or inhibi⁃tors have been identified so far,which can induce or inhibit ferroptosis through different targets and mecha⁃nisms,and are of potential clinical value in treating above-mentioned various diseases.This review summarizes the existing ferroptosis inducers and inhibitors in terms of structures,targets,features and appli⁃cation models in order to provide data for subsequent research and clinical application.
