茵陈草水提物对四氯化碳诱导的急性肝损伤小鼠的保护作用研究 被引量:4

摘要 目的:研究茵陈草水提物对四氯化碳诱导的急性肝损伤小鼠的保护作用机制。方法:采用四氯化碳诱导建立急性肝损伤小鼠模型。将成模小鼠随机分为模型组、水飞蓟素组、茵陈草水提物低、中、高剂量组,每组10只。连续灌胃给药56 d,每日1次。另设10只SD小鼠为空白组。末次给药后,试剂盒检测小鼠血清中AST、ALT的含量、肝组织锰-超氧化物歧化酶(Mn-SOD)、过氧化氢酶(CAT)的活性及活性氧自由基(ROS)、丙二醛(MDA)的含量。HE染色法观察小鼠肝脏的病理学变化。Western蛋白免疫印迹法检测肝脏兔抗小鼠转录激活因子4(ATF 4)、CCAAT/增强子结合蛋白同源蛋白(GHOP)的蛋白水平。结果:空白组、茵陈草水提物各浓度组小鼠血清AST、ALT、肝组织中MDA、ROS的含量及ATF4、GHOP的蛋白表达较模型组低,肝组织中Mn-SOD、CAT的活性较模型组高(P<0.05)。结论:茵陈草水提物对四氯化碳所致急性肝损伤小鼠的肝脏有一定保护作用,其作用机制可能与提高抗氧化因子活性,改善氧化应激损伤,抑制内质网应激通路中的相关蛋白有关。 Objective:To investigate the protective effect of water extract of Artemisia dracunculus L.in acute liver injury mice induced by carbon tetrachloride.Methods:A mouse model of acute liver injury was induced by the chemical poison carbon tetrachloride.The model mice were divided into model control group,silymarin group,and low,medium and high dose of Artemisia dracunculus L.water extract groups with 10 mice in each group.Mice were administered once a day with corresponding drug for 56 days.Ten healthy mice were in normal control group.After the last administration,the serum levels of aspartate aminotransferase(AST)and alanine aminotransferase(ALT)were detected by using commercial kits.The activities of manganese-superoxide dismutase(Mn-SOD),catalase(CAT),reactive oxygen species(ROS)and malondialdehyde(MDA)levels in liver were also determined.Western blot was used for the detection of ATF4 and GHOP.Results:Compared with those in the model control group,the serum contents of AST and ALT in the normal control group and Artemisia dracunculus L.water extract groups were decreased.The activities of Mn-SOD and CAT in liver tissue of the normal control group and Artemisia dracunculus L.water extract groups were increased,while the MDA and ROS levels were down-regulated(P<0.05).The protein expressions of ATF4 and GHOP in liver tissue of the normal control and Artemisia dracunculus L.water extract groups were lower than those in the model control group.Conclusion:The water extract of Artemisia dracunculus L.shows protective effect on carbon tetrachloride induced acute liver injury in mice,and the underlying mechanism may be associated with increasing the activity of antioxidant factors,improving oxidative stress damage,and inhibiting related proteins in endoplasmic reticulum stress pathway.
