摘要 采用高效液相离子交换法对电泳纯SIEA 28 kDa抗原组分进一步分离纯化。对获得的色谱纯SIEA 28 kDa抗原分子进行薄层等电聚焦分析,测定其等电点。结果证明电泳纯SIEA 28 kDa抗原分子经高效液相离子交换法纯化后,280nm、220nm波长色谱图均为一个两侧对称、光滑的高峰曲线,色谱洗脱液对其免疫活性光密度图也为一个两侧对称、光滑的高峰曲线。色谱纯化证明SIEA 28 kDa抗原组分可能为单一分子,其等电点范围在p15-p17之间。 The SIEA 28 kDa component of Schistosoma japonicum purified by gel electrophoresis was isolated and purified further by ion exchange high performance liquid chromatography (HPLC) and then its pI was measured by thin-layer PAG-IEF. After purified by HPLC, its component took the appearance of a symmetric smooth chromatography curve with a single peak both in 220 nm and 280 nm wavelength. The curve of chromatography elusion verse its activity was also symmetric and single-peaked. The result shows that SIEA 28 kDa component is probably a single molecule and its pI is ranged from 5 to 7.
