摘要
目的探讨微小RNA-153(miR-153)、吲哚胺-2,3-双加氧酶1(IDO1)在儿童急性白血病中的表达水平及临床意义。方法将本院血液科收治的急性白血病患儿42例作为研究对象,并选取同时间段内健康体检儿童作为对照。用实时荧光定量逆转录聚合酶链反应(qRT-PCR)技术检测所有研究对象外周血血浆miR-153、IDO1 mRNA表达水平;用间接免疫荧光法检测外周血单个核细胞IDO1蛋白表达水平;分析急性白血病患儿外周血血浆miR-153表达水平、外周血单个核细胞IDO1阳性表达与急性白血病临床特征的关系;采用Pearson法分析外周血血浆miR-153与IDO1相关性;用Kaplan-Meier生存曲线分析外周血血浆miR-153、外周血单个核细胞IDO1表达水平对急性白血病患儿预后影响。结果白血病组患儿、健康组儿童外周血血浆miR-153 mRNA相对表达量分别为0.54±0.18,1.03±0.24,外周血单个核细胞IDO1 mRNA水平分别为1.81±0.40,0.85±0.21,外周血单个核细胞IDO1蛋白阳性表达率分别为61.90%,0,差异均有统计学意义(均P<0.05);外周血血浆miR-153表达水平、外周血单个核细胞IDO1阳性表达与肺部是否有感染有关(P<0.05);外周血血浆miR-153表达水平与IDO1表达水平呈负相关(r=-0.731,P<0.05);急性白血病患儿外周血血浆miR-153低表达组、miR-153高表达组治疗后3年生存率分别为50.00%,82.14%,差异有统计学意义(P<0.05),外周血单个核细胞IDO1阳性组、IDO1阴性组治疗后3年生存率分别为57.69%,93.75%,差异有统计学意义(P<0.05)。结论急性白血病患儿外周血血浆miR-153下调、IDO1表达上调,外周血单个核细胞IDO1阳性表达率升高,miR-153表达水平、IDO1阳性表达与患儿预后密切相关。
Objective To investigate the expression levels of microRNA-153(miR-153)and indoleamine-2,3-dioxygenase-1(IDO1)in children with acute leukemia and their clinical significances.Methods Forty-two children with acute leukemia admitted to the Department of Hematology in our hospital were selected as the study subjects,and healthy children in the same period were selected as control group.Real-time quantitative polymerase chain reaction(qRT-PCR)was used to detect the expression levels of miR-153 and IDO1 mRNA in peripheral blood plasma of all subjects.The expression of IDO1 protein in peripheral blood mononuclear cells was detected by indirect immunofluorescence assay.The relationships between the expression of miR-153 in peripheral blood plasma,the positive expression of IDO1 in peripheral blood mononuclear cells and the clinical characteristics of acute leukemia in children were analyzed.Pearson method was used to analyze the correlation between miR-153 and IDO1 in peripheral blood plasma.Kaplan-Meier survival curve was used to analyze the prognostic effects of miR-153 in peripheral blood plasma and IDO1 expression in peripheral blood mononuclear cells on children with acute leukemia.Results The relative expression levels of peripheral blood plasma miR-153 mRNA in children with leukemia and healthy children were 0.54±0.18 and 1.03±0.24,the levels of IDO1 mRNA in peripheral blood mononuclear cells were 1.81±0.40 and 0.85±0.21,the positive expression rates of IDO1 protein in peripheral blood mononuclear cells were 61.90%,0,all with significant difference(all P<0.05).The expression level of miR-153 in peripheral blood plasma and IDO1 positive expression in peripheral blood mononuclear cells were related to pulmonary infection(P<0.05).The expression of miR-153 in peripheral blood plasma was negatively correlated with the expression of IDO1(r=0.731,P<0.05).The 3-year survival rates in the low the high miR-153 expression group after treatment were 50.00%and 82.14%respectively in peripheral blood of children with acute leukemia,while the 3-year survival rates in the IDO1 positive and negative group were 57.69%and 93.75%,respectively,and the differences of above data between the two groups were statistically significant(P<0.05).Conclusion The level of miR-153 in peripheral blood of children with acute leukemia is down-regulated,the expression of IDO1 is up-regulated,and the positive expression rate of IDO1 in peripheral blood mononuclear cells is increased.The expression levels of miR-153 and positive expression of IDO1 are closely related to the prognosis of children.
