摘要
目的研究CdSe/ZnS量子点对肺癌顺铂耐药株A549/cis凋亡的影响及作用机制。方法将A549/cis细胞分为空白组和低、中、高剂量实验组。空白组给予正常DMEM培养基进行培养;低、中、高剂量实验组分别给予200,400,600 nmol·L-1 CdSe/ZnS量子点100μL。用CCK-8法检测细胞增殖情况,用流式细胞术检测细胞凋亡率,用Western Blotting法检测凋亡蛋白和内质网应激蛋白的表达情况。结果CdSe/ZnS量子点对肺癌顺铂耐药株A549/cis的半数抑制浓度为453.69 nmol·L-1。低、高浓度实验组和空白组的凋亡率分别为(15.68±5.29)%,(50.36±6.79)%和(0.82±0.03)%,B淋巴细胞瘤-2蛋白分别为0.24±0.03,0.19±0.05和0.77±0.07,Bcl-2相关X蛋白分别为0.79±0.06,1.03±0.07和0.66±0.05,半胱氨酸天冬氨酸蛋白酶-8分别为0.08±0.02,0.25±0.04和0.04±0.01,葡萄糖调节蛋白-78分别为0.11±0.02,0.22±0.07和0.09±0.03,葡萄糖调节蛋白-94分别为0.16±0.07,0.35±0.04和0.15±0.05,高剂量实验组的上述指标与空白组比较,差异均有统计学意义(均P<0.05)。结论CdSe/ZnS量子点可以通过线粒体损伤途径和内质网途径抑制肺癌顺铂耐药株A549/cis细胞增殖,诱导其凋亡。
Objective To study the effect and mechanism of CdSe/ZnS quantum dots on apoptosis of cisplatin resistant lung cancer cell line A549/cis.Methods The A549/cis cells were divided into blank group and experimental-L,-M,-H groups.Blank group was treated with DMEM medium.Experimental-L,-M,-H groups were treated with 200,400,600 nmol·L-1 of CdSe/ZnS quantum dots of 100μL,respectively.The proliferation was detected using CCK-8 assay.Flow cytometry was used to detect the apoptosis rate.The expressions of apoptosis and endoplasmic reticulum stress proteins were measured using Western Blotting.Results The 50%inhibiting concentration of CdSe/ZnS quantum dots to A549/cis cells was 453.69 nmol·L-1.The apoptotic rates of experimental-L,experimental-H and blank groups were(15.68±5.29)%,(50.36±6.79)%and(0.82±0.03)%,the expression levels of B-cell lymphoma-2 protein were 0.24±0.03,0.19±0.05 and 0.77±0.07,the expression levels of Bcl-2 associated X protein were 0.79±0.06,1.03±0.07 and 0.66±0.05,the expression levels of cysteinyl aspartate specific proteinase 8 were 0.08±0.02,0.25±0.04 and 0.04±0.01,the expression levels of glucose-regulated protein 78 were 0.11±0.02,0.22±0.07 and 0.09±0.03,the expression levels of glucose-regulated protein 94 were 0.16±0.07,0.35±0.04 and 0.15±0.05.There were significant differences in the above-mentioned indexes between experimental-H group and blank group(all P<0.05).Conclusion CdSe/Zn S quantum dots induce the apoptosis of cisplatin resistant lung cancer A549/cis cells through the mitochondrial and endoplasmic reticulum pathway.
