脂肪来源干细胞通过降低TGF-β1和Ⅰ、Ⅲ型胶原的表达水平抑制增生性瘢痕成纤维细胞 被引量:3

摘要 目的 分析脂肪来源干细胞(adipose-derived stem cells, ADSCs)对增生性瘢痕成纤维细胞生长的影响。方法 对人脂肪组织进行剪碎、消化、过滤、培养传代后得到ADSCs,并进行鉴定;对增生性瘢痕组织进行剪碎、消化并培养传代得到增生性瘢痕来源的成纤维细胞;将ADSCs和增生性瘢痕来源的成纤维细胞共培养作为实验组;增生性瘢痕来源的成纤维细胞单独培养作为对照组。用ELISA检测两组成纤维细胞内的Ⅰ、Ⅲ型胶原蛋白的含量;Western blot法检测两组成纤维细胞Ⅰ、Ⅲ型胶原和α-SMA的表达;RT-PCR法检测两组的Ⅰ、Ⅲ型胶原和α-SMA相对表达量;采用Western blot和RT-PCR法检测两组细胞中TGF-β1的相对表达量。结果 实验组与对照组相比,TGF-β1、Ⅰ、Ⅲ型胶原和α-SMA的表达量均有明显下降,其差异具有统计学意义(P<0.05)。结论 ADSCs与增生性瘢痕成纤维细胞共培养后能减弱成纤维细胞的增殖能力,降低TGF-β1和Ⅰ、Ⅲ型胶原蛋白的表达,从而对增生性瘢痕起到抑制作用。 Objective To analyze the effect of adipose-derived stem cells(ADSCs)on the growth of hypertrophic scar fibroblasts.Methods The ADSCs were extracted from human adipose tissue and the fibroblasts were extracted from hypertrophic scar tissue through cutting,digestion and culture.The co-culture of ADSCs and fibroblasts was as the experimental group and the fibroblasts culture alone was as the control group.The content of collagenⅠandⅢin fibroblasts of both groups was detected by ELISA;the expression of collagenⅠ,Ⅲandα-SMA in fibroblasts of both groups was detected by Western blot and RT-PCR.The relative expression of TGF-β1 in fi-broblasts of both groups was detected by Western blot and RT-PC.Results The expression of TGF-β1,collagenⅠ,collagenⅢandα-SMA in the experimental group was significantly decreased compared with the control group.The difference was statistically signifi-cant(P<0.05).Conclusion The co-culture of ADSCs and hypertrophic scar fibroblasts could decrease the proliferation of fibroblasts and reduce the expression of TGF-β1,collagenⅠandⅢ,thus inhibiting the hypertrophic scar.
