摘要
目的探索miR-190a-5p对血管瘤内皮细胞生长、凋亡及转录因子4(TCF4)水平的影响.方法采用Lipofectamine^■2000构建miR-190a-5p、TCF4过表达EOMA细胞,应用BrdU染色、FCM法、荧光素酶试验、Western印迹检测增殖、凋亡、线粒体膜电位变化、验证miR-190a-5p与TCF4靶向关系.结果miR-190a-5p过表达可以抑制EOMA细胞增殖、促进细胞凋亡与线粒体膜电位下降,靶向作用TCF4,提高Bax/Bcl-2、Caspase-3、Caspase-9水平,降低c-Myc水平.结论miR-190a-5p过表达可对抑制血管瘤内皮细胞EOMA增殖、促进细胞凋亡,可能与靶向调控TCF4表达有关.
Objective To explore the effects of miR-190a-5p on growth and apoptosis of he-mangioma vascular endothelial cells(HVEC)and level of transcription factor4(TCF4).Methods The miR-190a-5p and EOMA of TCF4 overexpression were constructed by Lipofectamine^■.The proliferation,apoptosis and changes of mitochondrial membrane potential(MMP)were detected by BrdU staining,FCM and luciferase test.The targeted relationship between miR-190a 5p and TCF4 was validated by Western blotting.Results Overexpression of miR-190a 5p could inhibit prolifer-ation of EOMA cells,promote apoptosis and MMP decrease,target TCF4,increase levels of Bax/Bcl-2,Caspase-3 and Caspase-9,and reduce c-Mye level.Conclusion The overexpression of miR-190a-5p can inhibit proliferation of EOMA cells,and promote apoptosis,which may be related to targeting TCF4 expression.
