摘要
高敏感的检测试剂盒的基因检测,是防控新冠疫情的关键手段之一。鉴于目前国内外新型冠状病毒(SARS-CoV-2)检测试剂盒在敏感性方面存在的不足,我们认为与其所检测靶点数量的局限性有关。本研究结合生物信息学分析和采用重叠PCR合成策略,获得了可用于新型冠状病毒检测的8个新靶点。这些靶点主要呈现为两个方面的特点:一是增加了扩增引物的SARS-CoV-2特异性;二是增强了适宜于TaqMan探针检测的SARS-CoV-2特异性。这些新的靶点有望成为未来SARS-CoV-2高灵敏度检测及试剂盒开发的重要对象。
Detection kits with high sensibility are one of the key tools for COVID-19 control.The relatively low sensitivity of SARS-COV-2 detection kits presently used is partially related to the limited number of the viral gene fragments targeted.In this study,eight novel detection targets of SARS-COV-2 were obtained by combining bioinformatics analysis and overlapping PCR synthesis strategy.These targets are characterized by two aspects:SARS-COV-2 specificity of the amplified primers and being suitable for TaqMan probe detection.These new targets add important resource for high sensitivity detection and kit development of SARS-COV-2 in the future.
