摘要
获得一种解淀粉芽孢杆菌X8(Bacillus amylobacillus X8),其是一种抑制杀鲑气单胞菌D5-7(Aeromonas salmonicide D5-7)生长的拮抗菌,该菌在生长过程中菌液表面会形成一层细菌生物膜。为了探讨环境条件对B.amylobacillus X8生物膜形成的影响情况,通过打孔抑菌圈法探讨培养过夜的B.amylobacillus X8菌液在不同静置时间、静置温度、pH和金属离子存在下,对其生物膜形成的影响情况。结果表明,B.amylobacillus X8生物膜形成的最佳条件为:用pH=8的专用培养基过夜培养,然后在30℃下静置48 h,此时膜产量最高,且此条件下生物膜的抑菌活性最强;同时添加5 g/L K+时有助于膜的形成。该研究结果可为进一步研究B.amylobacillus X8拮抗菌生物膜的形成机制及抗菌机理奠定基础。
The research showed the Bacillus amylobacillus X8 was an antagonistic bacteria to Aeromonas salmonicide D5-7.It was surprised to find that the biofilm of B.amylobacillus X8 had formed on the surface of the cultivating liquid.In order to explore the influence of environmental conditions on the formation of B.amylobacillus X8 biofilm,the effects of different standing time,temperature,pH and metal ions on the biofilm formation of B.amyobacillus X8 were studied by means of perforated inhibition zone method.The results showed that the best conditions for the formation of B.amylobacillus X8 biofilm were as follows:overnight culture in special medium with pH=8,and then standing at 30℃for 48 h,the yield of biofilm was the highest,and the antibacterial activity of biofilm was the strongest under this condition.The addition of 5 g/L K+is helpful to the formation of the membrane.The results laid a foundation for further study on the formation mechanism and antibacterial mechanism of B.amylobacillus X8 antagonistic biofilm.
