Effect of a-2,3-sialyltransferase I on the biological behavior of prostate cancer PC3 cells

摘要 目的探讨α-2,3唾液酸转移酶Ⅰ(ST3Gal-Ⅰ)对PC3生物学行为的影响。方法将前列腺癌细胞系PC3分为空白对照组、阴性对照组、干预组。空白对照组不作任何处理,阴性对照组转染pEGFP-N1质粒,干预组转染pEGFP-N1-ST3Gal-Ⅰ质粒。采用Western blot检测三组中ST3Gal-Ⅰ蛋白表达量,采用CCK8法检测三组PC3增殖率,通过Transwell小室侵袭实验检测三组PC3侵袭能力,采用流式细胞术检测三组PC3凋亡率。结果干预组PC3增殖率、侵袭能力及ST3Gal-Ⅰ蛋白表达量明显高于空白对照组、阴性对照组(P<0.05);干预组PC3凋亡率明显低于空白对照组、阴性对照组(P<0.001);阴性对照组PC3增殖率、侵袭能力、凋亡率及T3Gal-I蛋白表达量与空白对照组比较,差异均无统计学意义(P>0.05)。结论 ST3Gal-Ⅰ能够促进PC3增殖、侵袭,抑制其凋亡。 Objective To investigate the effect of a-2,3-sialyltransferase I(ST3Gal-I)on the biological behavior of prostate cancer PC3 cells.Methods The prostate cancer PC3 cells were divided into blank control group,negative control group and intervention group.The blank control group received no treatment,the negative control group was transfected with pEGFP-Nl plasm id,and the intervention group was transfected with pEGFP-Nl-ST3Gal-I plasmid.The expression of ST3Gal-I protein,proliferation rate,invasion ability,and apoptosis rate of PC3 cells were detected by Western hlot,CCK8 method,Transwell chamber assay and flow cytometry Respectively.Results The proliferation rate,invasion ability and ST3Gal-I protein expression of PC3 cells in the intervention group were significantly higher than those in the blank control group and the negative control group(P<0.05).The apoptosis rate of PC3 cells in the intervention group was significantly lower than that in the blank control group and the negative control group(P<0.05).The proliferation rate,invasive ability ap optosis rate and ST3Gal-I protein expression of PC3 cells in the negative control group were not significantly different from those in the blank control group(P>0.05).Conclusions ST3Gal-I can promote the proliferation and invasion of prostate cancer PC3 cells and inhibit its apoptosis.
