一株柯萨奇病毒A组9型病毒的全基因组分析 被引量:1

The complete genome sequence analysis of a coxsackievirus A9 strain

摘要 目的对2010年云南省昆明市1例手足口病(hand,foot and mouth disease,HFMD)并发脑炎患儿的粪便中分离到的1株柯萨奇病毒A组9型(coxsackievirus A9,CV-A9)毒株的全基因组序列和遗传特性进行分析。方法利用人类横纹肌肉瘤(human rhabdomyosarcoma,RD)细胞对该粪便标本进行病毒分离,提取病毒RNA,采用反转录聚合酶链反应(reverse transcription-polymerase chain reaction,RTPCR)分段扩增病毒的全基因组序列,利用MEGA 6.06、Geneious 9.0.2和SimPlot 3.5.1等软件对该毒株的全基因序列进行分析。结果获得1株CV-A9分离株(A744/YN/CHN/2010,简称A744),其全基因组序列长为7319bp。该毒株与CV-A9重组株(A108/YN/CHN/2009)核苷酸一致性和氨基酸同源性最高,分别为97.4%和99.0%,可能发生重组事件。结论A744分离株为CV-A9,与中国其他CV-A9分离株同属D2基因亚型。 Objective To analyze the genome sequence and genetic characteristics of a coxsackievirus A9(CVA9)strain isolated from the feces of a child with viral encephalitis from hand,foot and mouth disease(HFMD)in Kunming,Yunnan province of China in 2010.Methods Virus strains in the fecal samples were isolated in RD cells,and the viral RNA was extracted.The whole genome sequence of the virus was amplified by RT-PCR.MEGA6.06,Geneious 9.0.2,SimPlot 3.5.1softwares were used to analyze the whole genome sequence.Results A CV-A9 isolate(A744/YN/CHN/2010,A744)was obtained,and its whole genome sequence was 7319bp.This strain had the highest homology of nucleotides and amino acids with the recombinant CV-A9strain(A108/YN/CHN/2009),which were 97.4%and 99.0%,respectively.Conclusions A744isolate is CV-A9,grouped as D2gene subtype with other CV-A9isolates in China.
