摘要
真菌非核糖体多肽通常具有多种生物学活性。本研究从球孢白僵菌基因组中挖掘出其非核糖体多肽合成酶基因(BbNRPS),克隆获得其全长c DNA,并对该基因进行生物信息学和表达谱分析。结果显示,BbNRPS基因cDNA开放阅读框总长4194 bp,编码1397个氨基酸残基,BLAST比对发现其包含A-T-C结构域;SignalP显示其为分泌蛋白,位于细胞质基质。系统发育显示BbNRPS蛋白序列与其他合成铁载体SidC的NRPS聚为一类,推测其合成铁载体。表达谱显示,肌糖能促进BbNRPS基因的表达。该研究为球孢白僵菌NRPS基因的挖掘提供参考。
Fungal nonribosomal peptides usually own a variety of biological activities.In this study,non-ribosomal polypeptide synthase gene(BbNRPS)was extracted from Beauveria bassiana genome,its full-length cDNA was cloned and analyzed with bioinformatic methods and expression spectrum analysis.The results showed that:the full length of BbNRPS open reading framer takes 4194 bp,and codes 1397 amino acids,BLAST alignment showed that BbNRPS have A-T-C three domains.It’s a signal peptide,locates in the cytoplasmic matrix.Phylogenetic analysis clarified that the BbNRPS was clustered in a branch with the other NRPS gene that synthesis the siderophore Sid C,speculating that it can synthesize SidC.The expression results showed that inositol could promote the expression of BbNRPS gene.The study provides a reference for the excavation of NRPS gene from Beauveria bassiana.
