摘要
为了探讨中等强度爬坡运动对肥胖大鼠AMPK-PGC-1α信号和脂代谢的影响,将80只雄性SD大鼠随机分为对照组、肥胖组、跑步组和爬坡组,对照组使用普通饲料喂养,肥胖组、跑步组和爬坡组使用高脂饲料喂养,共喂养6周。建模完成后,跑步组和爬坡组分别进行4周的跑步运动和爬坡运动。运动完成后检查各组大鼠的血糖、TC、TG、LDL-c、HDL-c、瘦素和脂联素水平。使用免疫组织化学染色、Western blotting或qRT-PCR检测大鼠骨骼肌组织中pAMPK、PGC-1α、MCAD、CPT-1、PPARα和PPARγ的蛋白或mRNA表达。结果显示,与跑步组比较,爬坡组的TC、TG和LDL-c水平均显著降低,而HDL-c显著升高;爬坡组的血清瘦素和脂联素含量均显著升高;爬坡组的pAMPK和PGC-1α蛋白表达水平均显著升高;爬坡组的MCAD、CPT-1、PPARα和PPARγmRNA表达水平均显著升高。研究表明,中等强度爬坡运动可通过促进瘦素和脂联素的合成分泌、激活AMPK-PGC-1α信号通路来改善糖脂代谢,从而发挥降血糖和降血脂的作用。
To explore the effects of moderate-intensity climbing exercise on AMPK-PGC-1αsignaling and lipid metabolism in obese rats,eighty male SD rats were randomly divided into the control group,obese group,running group and climbing group.The control group was fed with ordinary feed,and the obese group,running group and climbing group were fed with high-fat diet for 6 weeks.After the modeling was completed,the running group and the climbing group performed running and climbing exercises for 4 weeks,respectively.After exercise,the blood glucose,TC,TG,LDL-c,HDL-c,leptin and adiponectin levels were detected in each group of rats.Immunohistochemical staining,Western blotting or qRT-PCR was used to detect the protein or mRNA expression of pAMPK,PGC-1α,MCAD,CPT-1,PPARαand PPARγin rat skeletal muscle tissue.The results showed that compared with the running group,the levels of TC,TG and LDL-c in the climbing group were significantly reduced,while HDL-c was significantly increased;The serum leptin and adiponectin levels in the climbing group were significantly increased;The expression levels of pAMPK and PGC-1αprotein in the climbing group were significantly increased;The levels of MCAD,CPT-1,PPARαand PPARγmRNA in the climbing group were significantly increased.Studies have shown that moderate-intensity climbing exercises can improve the metabolism of glucose and lipids by promoting the synthesis and secretion of leptin and adiponectin,and activating the AMPK-PGC-1αsignaling pathway,thereby reducing blood glucose and lipids.
