摘要
从荸荠皮中分离出总皂苷并经AB-8大孔树脂纯化,在模拟胃液情况下,采用盐酸萘乙二胺法和α-萘胺法分别测定荸荠皮皂苷体外清除亚硝酸盐和阻断亚硝胺合成的能力,并比较不同模拟胃液、皂苷浓度、反应时间、反应体积和预处理温度对清除亚硝酸盐及阻断亚硝胺的影响。结果显示,荸荠皮皂苷在模拟胃液B环境下效果发挥更好,其对亚硝酸盐的清除率及亚硝胺的阻断率分别最高可达84.5%和75%,且IC50分别为6.02μg/mL和7.58μg/mL;随着反应浓度、反应时间和反应体积的增加,荸荠皮皂苷对亚硝酸盐清除作用与亚硝胺合成的阻断作用也逐步增强,在100℃温度处理后其活性也未受影响,热稳定性好。因此,荸荠皮皂苷具有良好的亚硝酸钠清除作用与阻断亚硝胺合成作用。
Total saponins were isolated from water chestnut peel and purified by AB-8 macroporous resin.The ability of water chestnut peel saponins to remove nitrite and block the synthesis of nitrosamine in vitro was determined by naphthalene ethylenediamine hydrochloride method andα-naphthaline method in simulated gastric juice.Compare the effect of different simulated gastric juice,saponin concentration,reaction time and volume on removing nitrite and blocking the synthesis of nitrosamines by water chestnut peel saponins.Water chestnut peel saponins played a better role in simulating gastric juice B.The results showed that the nitrite removal rate and nitrosamine blocking rate could reach as high as 84.5%and 75%separately;The IC50 was 6.02μg/mL and 7.58μg/mL,separately.It was indicated that the effect of nitrite and nitrosamine by water chestnut peel saponins gradually enhanced as saponin concentration,reaction time and volume increased.The water chestnut peel saponins exhibited good thermal stability as the treatment of 100℃,and the activity was not affected.Consequently,the water chestnut peel saponins had good effects on nitrite scavenging and blocking nitrosamine synthesis.
