摘要
提纯胎菊中绿原酸,建立一套高效的纯化工艺,并对产品进行结构鉴定。将胎菊粉碎,超声波提取得粗提液,经大孔树脂初步纯化,乙酸乙酯萃取,石油醚分相,聚酰胺柱色谱分离,乙酸乙酯重结晶得产品;经过高效液相色谱、紫外-可见光谱、傅里叶变换红外光谱、核磁共振氢谱技术分析得产品为绿原酸,产率为0.018%,经高效液相色谱定量,其纯度可达98.7%。采用超声波粗提-大孔树脂纯化-萃取分相-聚酰胺柱层析分离-产品结晶的工艺,可从胎菊中得到高纯度的绿原酸产品。
To purify chlorogenic acid from Foetus chrysanthemum,establish a set of efficient purification technology,and identify the product structure.The crude extract of Foetus chrysanthemum was extracted by ultrasonic wave,purified by macroporous resin,extracted by ethyl acetate,separated by petroleum ether,separated by polyamide column chromatography and recrystallized by ethyl acetate.The product was chlorogenic acid by HPLC,UV-Vis,FTIR and 1 H-NMR.The yield was 0.018%.The purity of HPLC was 98.7%.High purity chlorogenic acid can be obtained from Foetus chrysanthemum by ultrasonic extraction,macroporous resin purification,extraction and phase separation,polyamide column chromatography and product crystallization.
