摘要
目的运用FilmArray全自动病原微生物检测系统快速检测方法与国家标准方法对一份食品样本中食源性致病菌同时开展检测,探讨FilmArray在食品中食源性致病菌检测的应用。方法将1份生牛肉样品分别用E.coli肉汤(EC)和缓冲蛋白胨水(BPW)两种增菌液增菌后,使用FilmArray检测系统的GI Panel胃肠道感染测试条进行检测;按照GB 4789以及《霍乱防治手册》规定的方法对12种常见的食源性致病菌:沙门菌、弯曲菌、志贺菌、副溶血弧菌、霍乱弧菌、5种致泻性大肠埃希菌(EAEC、ETEC、EPEC、STEC、EIEC)、O157、小肠结肠炎耶尔森菌进行检测。结果运用FilmArray对食品样本经两种增菌液增菌后的检测结果一致,检出6种食源性致病菌,包括类志贺邻单胞菌、霍乱弧菌、沙门菌、STEC、ETEC、EIEC。按照国标以及《霍乱防治手册》方法检出5种食源性致病菌,包括霍乱弧菌(非O1/O139)、沙门菌(德尔卑)、STEC、ETEC、EIEC。结论两种方法检测出食品样本中的食源性致病菌基本一致,FilmArray可对食品样本增菌液进行快速初筛检测,为后续病原菌分离提供一定指示作用。
Objective To investigate the application of FilmArray in the detection of foodborne pathogens in food samples and the national standard method was used to test simultaneously.Methods After a raw beef sample was enriched with two enrichment solutions of E. coli broth(EC) and buffered peptone water(BPW), the FilmArray GI Panel was used to detect the pathogens. In accordance with GB 4789 and the Cholera Prevention and Control Manual,about 12 common foodborne pathogens were tested,which were including salmonella,campylobacter,shigella,vibrio parahaemolyticus,vibrio cholerae,the diarrheogenic Escherechia coli(EAEC,ETEC,EPEC,STEC, EIEC), O157, yersinia enterocolitis. Results The results of the FilmArray GI Panel were consistent with that of the two bacteria-increasing solutions in the food samples. Six foodborne pathogens were detected, including shigella,vibrio cholerae, salmonella, STEC, ETEC and EIEC. Five foodborne pathogens were detected in accordance with national standards and the cholera prevention and control manual, including vibrio cholerae(non-O1/O139), salmonella(S.Derby), STEC, ETEC and EIEC. Conclusion The detection of foodborne pathogenic bacteria in food samples by the two methods is basically the same. FilmArray GI Panel can be used for rapid initial screening and detection of the bacteria solution in food samples, which can provide certain indication for the subsequent pathogen separation.
