摘要
采用高效液相色谱-串联质谱技术(HPLC-MS/MS),对不同类型番茄果实的叶酸及其衍生物含量进行初步分析。结果表明,栽培种和野生种不同基因型番茄果实中总叶酸含量均差异显著,栽培种番茄总叶酸含量为1532.79~4530.72μg·kg^-1,野生种番茄总叶酸含量为2587.85~4035.93μg·kg^-1。基于全基因组数据选取的17份醋栗番茄总叶酸含量为1934.81~5050.13μg·kg^-1。栽培种M82和野生种LA0443的叶酸含量较高,分别为4530.72μg·kg^-1和5050.13μg·kg^-1。对12种叶酸衍生物的分析结果表明,不同基因型番茄的叶酸衍生物种类也存在一定差异,大部分资源至少含有9种叶酸衍生物。其中醋栗番茄资源叶酸变异更为丰富:5份资源具有12种叶酸衍生物,而四氢叶酸(THF)、四谷氨酸5-甲酰四氢叶酸(5-F-THFGlu4)、叶酸(FA)、10-甲酰叶酸(10-F-FA)等在一些资源中含量较低或未能检测到。总体而言,番茄与大多数作物类似,5-甲基四氢叶酸(5-M-THF)是果实中的主要叶酸形式,其次是5-甲酰四氢叶酸(5-F-THF),二者呈现显著负相关。
Adopting high-performance liquid chromatograph coupled with tandem mass spectrometry(HPLC-MS/MS),this study conducted initial analysis on various forms of folic aid from different tomato species and its derivative contents.The results showed that there were distinguished differences in total folic acid contents in tomato fruits with different genotypes from cultispecies and wild species.The total folic acid contents of tomato cultispecies was 1532.79-4530.72μg·kg^-1 and that in wild species was 2587.85-4035.93μg·kg^-1.The total folic acid contents of 17 Solanum pimpinellifolium accessions selected based on whole genome sequence was 1934.81-5050.13μg·kg^-1.Thereinto,the folic acid contents of cultispecies M82 and wild species LA0443 were higher,being 4530.72μg·kg^-1 and 5050.13μg·kg^-1,respectively.The results of analyzing 12 kinds folic acid derivatives indicated that there were certain differences existing among folic acid derivatives types from different germplasm resources,most of them containing at least 9 folic acid derivatives.And the folic acid variation of these 17 S.pimpinellifolium accessions were more rich:5 accessions had 12 folic acid derivatives.While,the contents of tetrahydrofolate(THF),tetraglutamyl 5-formyltetrahydrofolate(5-F-THFGlu4),folic acid(FA)and 10-formyl folic acid(10-F-FA)were lower or even undetectable in some germplasm resources.In general,tomato was similar with the most crops,5-methyl-tetrahydrofolate(5-M-THF)was the major form of folic acid in fruit,followed by 5-formyltetrahydrofolate(5-F-THF).Moreover,a significant negative correlation was observed between 5-M-THF and 5-F-THF.
