摘要
目的建立测定罗汉松种子总黄酮含量的紫外-可见分光光度法。方法采用70%乙醇回流提取罗汉松种子中的总黄酮,以芦丁为对照品,经显色后,以紫外-可见分光光度计于253 nm波长处测定总黄酮含量。结果芦丁质量浓度在0.639~1.758μg/m L范围内与吸光度线性关系良好,线性回归方程为Y=0.904 4 X+1.073 3(r=0.997 3);平均加样回收率为101.60%,RSD为2.34%(n=6);罗汉松种子中总黄酮平均含量为4.66%。结论该方法操作简便,重复性好,适用于测定罗汉松种子中的总黄酮含量。
Objective To establish an ultraviolet-visible spectrophotometry method for the content determination of total flavonoids in the seed of Podocarpus macrophyllus.Methods Total flavonoids were extracted in the seed of Podocarpus macrophyllus by reflux method with 70%ethanol as extraction solvent.The rutin was used as the reference substance,and the content of total flavonoids was detected by ultraviolet-visible spectrophotometry at 253 nm after the samples were coloured.Results Rutin showed a good linear relationship in the range of 0.639-1.758μg/mL and the linear regression equation was as follows:Y=0.9044 X+1.0733(r=0.9973).The average recovery was 101.60%and RSD was 2.34%(n=6).The average content of total flavonoids in the seed of Podocarpus macrophyllus was 4.66%.Conclusion The method is simple and reproducible,which is suitable for the content determination of total flavonoids in the seed of Podocarpus macrophyllus.
