摘要
糖尿病的患病率逐年增加,给病人带来巨大的痛苦和经济负担。糖尿病的主要危害在于其并发症,尤其是心血管并发症,是导致死亡的主要原因。内皮细胞功能障碍是发生糖尿病血管并发症的重要因素之一。硫化氢(hydrogen sulfide,H2S)是继一氧化氮(nitric oxide,NO)和一氧化碳(carbon monoxide,CO)之后的第3种气体信号分子,发挥重要的生理与病理生理学效应。目前研究发现,H2S在糖尿病及糖尿病并发症中发挥着重要的保护作用,这已成为国内外研究的热点。H2S的生物利用障碍被认为是内皮细胞炎症及其相关的内皮功能障碍的新机制。H2S通过抑制内皮依赖性舒张反应受损、炎症反应、氧化应激、细胞凋亡、细胞自噬等减轻糖尿病血管内皮细胞功能障碍。本篇综述就H2S在糖尿病血管内皮细胞功能障碍中的作用及机制研究进展进行阐述。
The prevalence of diabetes has increased year by year,bringing huge pain and economic burden to patients.The main harm of diabetes is its complications,especially cardiovascular disease,which is the main cause of death.Endothelial dysfunction is an important factor in the occurrence of diabetic vascular complications.Hydrogen sulfide(H2S),the third endogenous gaseous molecule identified after nitric oxide(NO)and carbon monoxide(CO),plays an important role in both physiology and pathophysiology.Recently,studies found that H2S plays a vital protective role in the pathogenesis of diabetes and its complication,and become one of the hottest research topics.Disrupted H2S bioavailability is postulated to be a new mechanism for endothelial cell inflammation and its associated endothelial dysfunction.H2S can attenuate diabetic endothelial dysfunction through a variety of ways,including the inhibition of endothelium-dependent vasodilatation,inflammation,oxidative stress,apoptosis,autophagy and so on.In this review,we will summarize recent advances about the roles and mechanisms of H2S in the pathogenesis of diabetic vascular endothelial dysfunction.
